摘 要:[目的]研究CyclinE、CDK4、VEGF、c-myc及p53基因在肝癌中的表达及其意义。[方法]采用荧光实时定量PCR技术、免疫组织化学技术及蛋白免疫印迹技术对肝癌组织、自身癌旁组织的CyclinE、CDK4、VEGF、c-myc及p53基因的表达水平进行检测。[结果]肝癌组织中的CyclinE、CDK4、VEGF、c-myc及p53 m RNA的表达水平均高于自身癌旁组织(P均<0.05);CyclinE蛋白在肝癌组织中的阳性表达率为59.52%;CDK4蛋白在肝癌组织中的阳性表达率为40.48%;VEGF蛋白在肝癌组织中的阳性表达率为35.71%;c-myc蛋白在肝癌组织中的阳性表达率为7.62%;p53蛋白在肝癌组织中的阳性表达率为52.38%;而在癌旁组织不表达或呈微弱表达。相比于中、高分化肝癌患者,低分化癌组织中的各蛋白的阳性表达率均显著升高(CyclinE:94.74%vs 30.43%;CDK4:47.37%vs 34.78%;VEGF:42.11%vs 30.43%;c-myc:57.89%vs 39.13%;p53:63.16%vs 43.48%),且差异有统计学意义(均P<0.05)。相比于没有发生肝外转移的肝癌患者,有肝外转移的癌组织中的各蛋白的阳性表达率均显著升高(CyclinE:81.25%vs 46.15%;CDK4:68.75%vs 23.08%;VEGF:43.75%vs 30.77%;c-myc:62.50%vs38.46%;p53:62.50%vs 46.15%),且差异有统计学意义(P均<0.05)。[结论 ]CyclinE、CDK4、VEGF、c-myc及p53基因在人肝细胞癌组织中的表达呈阳性,且阳性表达程度与肿瘤分化程度和有无肝外转移相关。[Purpose] To investigate the expression and medical significance of CyclinE,CDK4,VEGF,c-myc and p53 gene in hepatocellular carcinoma(HCC). [Methods] Genetic levels of these genes in HCC and adjacent tissues were detected by quantitative real-time PCR(Q-PCR),and the protein expression levels were detected by Western Blot and immunohistochemistry. [Results]Each of those genes expression in HCC tissues was significantly higher than that in adjacent tissues(P〈0.05). The positive expression rates of CyclinE,CDK4,VEGF,c-myc and p53 were 59.52%,40.48%,35.71%,7.62% and 52.3%,respectively. The positive expression rate of each gene in poorly differentiated hepatocellular carcinoma was significantly higher than that in moderately and welldifferentiated carcinoma(CyclinE:94.74% vs 30.43%;CDK4:47.37% vs 34.78%;VEGF:42.11% vs30.43%;c-myc:57.89% vs 39.13%;p53:63.16% vs 43.48%,P〈0.05). And the positive expression of each gene was also significantly higher in the tissues of hepatocellular carcinoma metastasis than that in tissues without metastasis( CyclinE:81.25% vs 46.15%;CDK4:68.75% vs 23.08%;VEGF:43.75% vs 30.77%;c-myc:62.50% vs 38.46%;p53:62.50% vs 46.15%,P〈0.05). [Conclusion] CyclinE,CDK4,VEGF,c-myc and p53 genes are highly expressed in tissues of HCC both in genetic and protein level and correlated with tumor differentiation and metastasis.
