摘  要:目的研究6-姜酚对β淀粉样蛋白(Aβ_(1-42))引起的PC12细胞损伤的影响及可能的作用机制。方法 PC12细胞接种于含50ng/ml神经生长因子的F12K无血清培养液中,经5d诱导分化后,将细胞分为5组:对照组、Aβ_(1-42)干扰组,分别以6-姜酚(80、120及200μmol/L)进行预处理4h后的6-姜酚低、中、高剂量预处理组,再加入10μmol/L Aβ_(1-42)继续培养48h,通过Hoechst 33258核染色观察凋亡的细胞核形态改变,2',7'-二氯荧光黄双乙酸盐荧光探针法检测细胞内活性氧含量,微板法测定细胞内丙二醛及NO水平。结果与Aβ_(1-42)干扰组比较,6-姜酚低、中、高剂量预处理组凋亡细胞明显减少。Aβ_(1-42)干扰组较对照组显著增加活性氧(20.87±2.65 vs 2.40±0.25)、丙二醛[(0.89±0.09)nmol/mg vs(0.55±0.04)nmol/mg]及NO[(6.23±0.35)μmol/L vs(2.58±0.53)μmol/L,P<0.05]。与Aβ_(1-42)干扰组比较,6-姜酚低、中、高剂量预处理组活性氧、丙二醛和NO含量均降低,以6-姜酚中剂量预处理组降低最明显,分别为3.34±0.23,(0.69±0.07)nmol/mg和(2.95±0.28)μmol/L,差异有统计学意义(P<0.05)。结论 6-姜酚能抑制Aβ_(1-42)诱导的细胞凋亡,可能与其抗氧化及抗炎症作用有关。Objective To study the protective effect of 6-gingerol on amyloidβprotein(Aβ1-42-induced PC12 cell damage and its underlying mechanisms.Methods PC12 cells were seeded in a serum free F12 K medium containing 50ng/ml nerve growth factor(NGF).The cells were divided into control group,Aβ1-42intervention group,low 6-gingerol dose group,medium 6-gingerol dose group and high 6-gingerol dose group after they were cultured for 5days.The morphology of apoptotic nuclei was observed with Hoechst 33258 nuclear staining after the cells were cultured for48 h.Intracellular reactive oxygen species(ROS)level was measured using the DCFH-DA method,and intracellular malondialdehyde(MDA)and nitric oxide(NO)levels were measured with an assay kit.Results The Aβ1-42induced apoptosis was significantly lower in 6-gingerol intervention group than in Aβ1-42intervention group.The intracellular levels of ROS,MDA and NO were significantly higher in Aβ1-42intervention group than in low 6-gingerol dose group,medium 6-gingerol dose group and high 6-gingerol dose group(P〈0.05).Conclusion 6-gingerol inhibits Aβ1-42-induced apoptosis by reducing oxidative stress and inflammatory response.
