摘 要:目的探索阿里红多糖对卡氏肺孢子菌肺炎大鼠外周血清中TNF-α、s ICAM和IL-8 3种细胞因子含量及大鼠脾脏T淋巴细胞亚型分布的影响。方法建立SD大鼠PCP动物模型并随机分为5组,设立PCP模型对照组及阿里红多糖(Fomes Officinalis Polysaccharides,FOP)低(14 mg/kg)、中(28 mg/kg)、高剂量(56 mg/kg)治疗组,以复方新诺明作为阳性药物对照组,并设立空白SD大鼠为正常对照。ELISA检测各实验组大鼠外周血清中TNF-α、s ICAM-1和IL-8 3种细胞因子含量,流式细胞术分别检测PCP模型组和高剂量FOP组大鼠脾脏T淋巴细胞中CD4+T和CD8+T细胞百分比。结果各剂量阿里红多糖均能显著下调PCP大鼠外周血清中TNF-α、s ICAM-1的表达,且下调作用随FOP剂量增加而加大,组间结果比较差异有统计学意义(TNF-α:F=3 567.47,P<0.01;s ICAM-1:F=5 845.48,P<0.01);低剂量FOP组大鼠外周血清中IL-8含量高于PCP模型组,中剂量FOP组大鼠外周血清中IL-8含量低于PCP模型组(F=6.971,P<0.01),高剂量FOP组大鼠外周血清中IL-8含量与PCP模型组相比显著下降,但与SMZ-TMP阳性药物对照组相比差异无统计学意义(F=0.003,P>0.05);高剂量FOP组大鼠脾脏T细胞亚群分布中CD4+T细胞百分比显著增高,与PCP模型组大鼠相比差异有统计学意义(F=903.316,P<0.01)。结论阿里红多糖对PCP的治疗机制主要与其对机体免疫功能的调节作用有关,可能通过上调CD4+T细胞百分比和下调机体外周血清中PCP发生相关炎性介质的表达有关。Objective To study the effect of Fomes Officinalis Polysaccharides on the contents of TNF- α、s ICAM- 1、IL- 8 in peripheral serum and the percentages of T cells in Splenic of rats with Pneumocystis carinii pneumonia. Methods PCP rat models were established and then divided the model into five groups randomly: PCP model control group,Low dose Fo P group( 14 mg / kg),Moderate dose Fo P group( 28 mg / kg),High dose Fo P group( 56 mg / kg),Cotrimoxazole control group. At the same time normal control group was established. The contents of TNF- α、s ICAM- 1、IL- 8 in peripheral serum of rats in each group were detected by ELISA,and the percentages of CD4+T cells and CD8+T in Splenic of rats in PCP model control group and High dose Fo P group were detected by flow cytometry( FCM). Results Each dose of FOP could significantly downregulated the expression of TNF- α and s ICAM- 1 in peripheral serum of rats with PCP( TNF- α: F = 3 567. 47,P 〈0. 01; s ICAM- 1: F = 5 845. 48,P 〈0. 01),and the downregulation increased with the increase of dose. The contents of IL- 8 in peripheral serum of rats in Low dose Fo P group were higher than the PCP model control group,weal lower in the Moderate dose Fo P group( F =6. 971,P〈 0. 01). There were no significant differences compared with the Cotrimoxazole control group( F = 0. 003,P〈 0. 05),the High dose Fo P could significantly downregulated the expression of IL- 8 in peripheral serum of rats with PCP,and also could increase the percentages of CD4+T cells in Splenic of PCP rats( F = 903. 316,P 〈0. 01).
