摘  要:目的分离并筛选生姜抗化疗呕吐的有效部位,并对其抗化疗呕吐的作用机制作初步研究。方法以经典的化学提取分离方法分离生姜的不同提取部位,以小鼠胃排空模型及大鼠异食癖模型对生姜挥发油组分、生姜水提醇沉上清液部分、生姜水提醇沉沉淀部分及二次分离生姜水提醇沉上清液部分得到的生姜脂溶性部位和水溶性部位进行研究分析,确定生姜抗化疗呕吐的有效部位;采用离体豚鼠回肠实验,观察生姜有效部位提取物对5-HT引起的豚鼠回肠收缩的影响。结果生姜脂溶性部位能够显著的延缓顺铂引起的小鼠胃排空(P﹤0.05),并且减少顺铂引起的大鼠摄食高岭土的量(P﹤0.05);生姜脂溶性部位能够拮抗5-HT引起的离体豚鼠回肠收缩(P﹤0.05)。结论生姜脂溶性部位是其抗化疗呕吐的有效部位,其抗化疗呕吐的作用机制可能与其能够拮抗5-HT受体有关。Objective To separate and screen the anti- CINV active extract of Ginger and observe the effect of active extract of Ginger on the contraction of isolated ileum of guinea pig in vitro. Methods Gastric emptying in cisplatin- treated mice and pica mode of rats were applied to select the active anti- CINV extract of Ginger; and 5- HT was used to observe the contraction of ileum of guinea pig in vitro. Results The cisplatin- induced delay in gastric emptying of mice was ameliorated and the kaolin intake of rats was decreased significantly by the fat- soluble extract; meanwhile the contraction induced by 5- HT was inhibited on the isolated ileum of guinea pig in vitro. Conclusion The fat- soluble extract of Ginger was the anti- CINV active extract of Ginger and the anti- CINV mechanism of Ginger may be related to it's antagonism on 5- HT.
