摘 要:目的建立天然沉香中白木香酸含量的分析方法,研究白木香酸与浸出物含量(%)之间的相关性。方法按《中国药典》(2010年版)一部进行醇溶性浸出物的含量测定和理化鉴别。高效液相色谱法采用CNW Athena C_(18)-WP柱(250mm×4.6mm,5μm),以乙腈-0.1%(体积分数)磷酸水溶液(40∶60)为流动相,流速1.0ml·min^(-1),检测波长217nm,柱温35℃。结果白木香酸在2.000~80.00μg·ml^(-1)(r=0.9999)范围内线性关系良好,平均回收率为96.1%,RSD为3.0%(n=6)。在理化鉴别和浸出物含量合格的情况下,天然沉香中白木香酸与浸出物含量之间呈正比关系。结论高效液相色谱法可准确测定天然沉香中白木香酸的含量,结合白木香酸与浸出物含量之间的相关性,为天然沉香的质量评价提供依据。Objective To establish a HPLC method for determination of agaropiric acid in natural agarwood and research the relativity of contents between the ethanol- soluble extraction and agaropiric acid. Methods According to those correlative standards of Aquilariae Lignum Resinatum in Ch. P.( 2010 Edith),the content of ethanol- soluble extraction were determined and the physicochemical properties of samples were identified. The determination was performed on a CNW Athena C_(18)- WP column( 250 mm × 4. 6 mm,5μm) and the mobile phase consisted of acetonitrile- 0. 1% phosphoric acid( 40: 60). The flow rate was1. 0 ml·min^(-1). The detection wave length was 217 nm and the column temperature was 35℃. Results The good linear range of agaropiric acid was 2. 000 ~ 80. 00 g·ml^(-1)( r = 0. 9999). The average recovery was 96. 1% with RSD = 3. 0%( n = 6). Under the circumstance that the content of ethanol- soluble extraction and the physicochemical properties of samples were qualified,the relation of contents between the ethanol- soluble extraction and agaropiric acid of natural agarwoods was directly proportional.Conclusion This method is accurate based on HPLC determination of agaropiric acid. The relativity of contents between the ethanol- soluble extraction and agaropiric acid provided a reference to quality evaluation of natural agarwood.
