摘  要:【目的】研究葛根素对人成骨样MG-63细胞增殖的影响,并初步探讨其相关的分子作用机制。【方法】在前期研究工作的基础上,以同时表达两种雌激素受体(estrogen receptor,ER)亚型的人成骨样MG-63细胞为研究模型,通过RT-PCR、Western blotting及小RNA干扰等技术,探讨葛根素对人成骨细胞增殖的作用及作用机制。【结果】葛根素可显著促进MG-63细胞增殖,进一步研究发现葛根素可通过增加MG-63细胞Cyclin B1,Cyclin D1分子的表达水平使G1期细胞比例减少,G2+S期细胞比例增加。葛根素的上述作用能被纯ER阻断剂ICI 182,780部分阻断,利用小RNA干扰技术证实葛根素的促增殖作用是依赖ERα和ERβ的。【结论】葛根素对成骨细胞增殖作用的调节是通过ER途径,由ERα和ERβ共同介导的。【Objective】To study the effect of puerarin on human osteoblast-like MG-63 cell proliferation, and preliminarily explore its related molecular mechanism.【Methods】On the basis of previous research, human osteoblast-like MG-63 cells which expressed two estrogen receptor(ER) isoforms simultaneously were used as models. The effect of puerarin on human osteoblast proliferation and its mechanism were explored with RT- PCR, Western blotting and small RNA interference technology. 【Results】 Puerarin could significantly promote the MG-63 cell proliferation. Further research showed that puerarin could reduce G1 phase cell proportion and increase G2 + S phase cell percentage by improving the expression of Cyclin B1 and Cyclin D1 in MG-63 cells. Treatment with the abrogates The above effects of puerarin could be partially inhibited by ICI 182 and 780 which were ER antagonists. The stimulative effect of puerarin on proliferation mediated by both ERα and ERβ was confirmed with si RNA technology.【Conclusion】Puerarin affects osteoblast proliferation through the ER pathway which is mediated via both ERα and ERβ.
