摘  要:目的:研究多聚左旋精氨酸(PLA)和脂多糖(LPS)协同诱导气道上皮细胞NCI-H292分泌白介素-6(IL-6)和白介素-8(IL-8)的信号通路机制。方法:将NCI-H292细胞分为空白组、PLA组、LPS组、PLA+LPS组,采用Western Blot检测各组JNK的磷酸化水平,并比较SP-600125作用与否对JNK磷酸化和IL-6、IL-8表达量的影响。结果 :PLA、LPS诱导NCI-H292 JNK磷酸化水平增加(P<0.05),两者有协同作用(P<0.01);LPS协同PLA诱导NCI-H292分泌IL-6、IL-8(P<0.05);SP-600125抑制PLA、LPS、PLA+LPS诱导的JNK磷酸化(P<0.05)和IL-6、IL-8分泌(P<0.05)。结论:JNK信号通路参与LPS协同PLA诱导NCIH292分泌IL-6和IL-8的过程。Objective To study the mechanism of signaling pathway through which PLA(Poly-L-arginine)and(lipopolysaccharide, LPS) synergistically activate the release of(interleukin-6, IL-6) and(interleukin-8, IL-8)in NCI-H292 cells. Methods NCI-H292 cells were grouped into control group, PLA group, LPS group and PLA +LPS group and the phosphorylation of JNK in each group was measured by Western blot. The IL-6 and IL-8 were detected by(enzyme-linked immunosorbent assay, ELISA). Results PLA and LPS increased the phosphorylation of JNK in NCI-H292 cells(P 〈0.05), and there was a synergistic effect between PLA and LPS(P 〈0.01). PLA and LPS synergistically induced the release of IL-6 and IL-8 in NCI-H292 cells(P 〈0.05). JNK was significantly inhibited by SP-600125 which was a specific inhibitor of JNK signaling(P 〈0.05), and the expression of IL-6 and IL-8 stimulated by PLA, LPS and PLA + LPS decreased statistically by SP-600125(P 〈0.05). Conclusion JNK signal pathway participates in the release of IL-6 and IL-8 that is stimulated by the synergism of PLA and LPS in NCI-H292 cells.
