诺氟沙星在日本鳗鲡体内的代谢动力学及残留消除规律

摘　　要：为研究诺氟沙星（NFX）在鳗鲡体内的代谢和消除规律，以超高效液相色谱-串联质谱法测定日本鳗鲡在混饲口灌后血液和组织中NFX的含量变化，并进行药动学分析。结果表明，NFX以30mg／kg的剂量单次混饲口灌日本鳗鲡后，吸收分布迅速，达峰时间（Tmax）、吸收（T1/2ke）和分布半衰期（T1/2a）分别为3．000、1．012和1．570h；NFX在鳗鲡体内消除较快，消除半衰期（正，加）为15．267h，总清除率（CL）为1．315I／（h·kg）。此外，峰浓度（Cmax）为1．273mg／L，药时曲线下面积（AUC0-∞）为22．670mg／（L·h）。NFX以30mg／kg的剂量连续3d混饲口灌日本鳗鲡后，在肌肉、肝脏、肾脏和血浆中的消除速率常数分别为0．144、0．125、0．102和0．0931／d。根据WT1.4计算的理论休药期（WDT）分别为肌肉22．97d，肝脏21．30d，肾脏33．40d，血浆18．29d。本研究结果为诺氟沙星在水产动物中的实际应用提供理论依据。To investigate the pharmacokinetics of Norfloxacin( NFX) in eel( Anguilla japonica),the concentration-time data of NFX was determined by UPLC- MS / MS. Pharmacokinetic analysis showed that after oral administration of mixed feed at the dose of 30 mg / kg,the absorption and distribution of NFX were fast,with the peak time( T_(max)),absorption( T_(1/2Ka)) and distribution half- life( T_(1 /2α)) of 3. 000,1. 012 and 1. 570 h,respectively. The elimination of NFX was also quick,with the elimination half- life( T_(1 /2)) of 15. 267 h and total body clearance( CL) was1. 315 L /( h·kg). In addition,peak concentration( C_(max)) was 1. 273 mg /L,and the area under the concentration- time curve( AUC_(0 ~ ∞)) was 22. 67 mg /( L · h). After 3 d of successive oral administration at the dose of 30 mg /( kg·d) NFX,the elimination rate constant were 0. 144,0. 125,0. 102 and 0. 093 1 / d in muscle,liver,kidney and plasma,respectively. In theory,the withdrawal time calculated by WT1. 4 program was 22. 97 d in muscle,21. 30 d in liver,33. 40 d in kidney and 18. 29 d in plasma. The results provided a theoretical basis for practical applications of norfloxacin in aquatic animals.
