涤痰汤对动脉粥样硬化兔血脂、氧化应激以及炎性因子的影响

摘　　要：目的观察涤痰汤对动脉粥样硬化兔血脂、氧化应激及炎性因子的影响,以期了解涤痰汤稳定动脉粥样硬化的机制。方法 48只健康雄性新西兰白兔随机分为空白组、动脉粥样硬化模型组、阿托伐他汀钙组和涤痰汤低、中、高剂量组,每组8只。空白组饲以普通饲料,其余各组饲以高脂饲料。8周后,随机选取动脉粥样硬化模型组兔2只查血脂,提示血脂明显增高;主动脉分离后制备HE片,可见明显的粥样斑块,提示动脉粥样硬化造模成功。其余各组动物在高脂饮食的基础上,每天给予相应的药物进行干预,连续喂养4周。检测兔血清血脂、SOD、MDA、TNF-α、IL-6、ICAM-1、VCAM-1,并制备主动脉HE片。结果与动脉粥样硬化模型组比较,涤痰汤能降低动脉粥样硬化兔血清TC、LDL-c,升高HDL-c,能提高SOD活性,降低MDA、TNF-α、IL-6、ICAM-1、VCAM-1值,主动脉组织形态学检查显示:涤痰汤高剂量组动脉粥样硬化改善最明显。结论涤痰汤具有较好的调血脂作用,同时能提高抗氧化应激能力,具有降低致动脉粥样硬化炎性因子的作用,其动脉粥样硬化形态学改变可能与上述改变相关。Objective To observe the effect of Ditan decoction on blood lipid,oxidant stress and inflammatory factor of atherosclerosis rabbits,and explore the mechanism of action of Ditan decoction on atherosclerosis. Methods 48 healthy male New Zealand white rabbits were randomly divided into blank group,atherosclerosis model group,low,medium and high dose of Ditan decoction groups and atorvastatin group,8 rabbits in each group. The rabbits in blank group were fed with common forage; the rest were fed with high lipid forage. After 8 weeks,2rabbits of atherosclerosis group were randomly selected to check blood lipid shown increased obviously,and atherosclerotic plaque was found in aorta to verify whether the model was completed. The animals of the rest groups were intervented with corresponding medicines on the basis of high lipid forage for 4 successive weeks. To observe the levels of blood lipid,SOD,MDA,TNF- α,IL- 6,ICAM- 1,VCAM- 1 and prepare aortic HE slices. Results Ditan decoction with different doses could decrease the levels of Tc,LDL- c,and increase HDL- c level,SOD activity and reduce MDA,TNF- α,IL- 6,ICAM- 1 and VCAM- 1. The morphology of aorta tissue showed that the atherosclerosis of the high dose group was the most obvious. Conclusions Ditan decoction can modulate the lipid metabolism,improve the ability of resistance to oxidant stress and reduce the inflammatory factors of atherosclerosis.
