摘  要:以‘秦美’猕猴桃无菌苗叶片为外植体,通过不定芽诱导、继代增殖和生根培养基的筛选,建立高频直接再生体系.结果表明,叶片出芽最佳培养基为MS+5.0mg/L 6-BA+0.2mg/L NAA,40d出芽率达100%,每个叶盘平均出芽7.4个;不定芽继代增殖最佳培养基为MS+5.0mg/L 6-BA+0.2mg/L NAA+0.1mg/L GA3,每30d继代1次,1~5代平均繁殖系数达6.43;不定芽生根最佳培养基为1/2 MS+0.7 mg/L IBA,30d生根率为91.11%.在土壤营养钵中,试管苗30d移栽成活率达92.47%.本试验成功建立了‘秦美’猕猴桃的叶片高频直接再生体系,为其遗传转化奠定了基础.In order to establish a high-frequency and direct regeneration system for‘Qinmei'kiwifruit(Actinidia deliciosa)and lay a foundation for its genetic transformation,the leaves of this cultivar were used as explants to establish the regeneration system through adventitious bud induction,proliferation and rooting.The results showed that the maximum regeneration frequency of adventitious buds was as high as100%,and one leaf disc,on the average,produced 7.4adventitious buds on the medium MS+5.0mg/L6-BA+0.2mg/L NAA.The optimum medium for adventitious bud multiplication was MS+5.0mg/L 6-BA+0.2mg/L NAA+0.1mg/L GA3,with an average multiplication coefficient of 6.43 from the first to the sixth generation.1/2MS+0.7mg/L IBA was the most suitable for the rooting of the regenerated adventitious buds,the rooting rate being 91.11%.Having transplanted into soil for 30 days,92.47% of the regenerated plantlets survived.
