南方红豆杉水提物联合顺铂对人肺癌A549细胞增殖抑制作用的研究

摘　　要：目的:研究南方红豆杉水提物联合顺铂对肺癌A549细胞增殖的抑制作用和机制。方法:将A549细胞分为不同浓度顺铂组、不同浓度南方红豆杉水提物组及空白组,用CCK-8法检测药物作用48 h后对细胞存活率的影响。根据上述结果,再将A549细胞分为低浓度顺铂联合不同浓度南方红豆杉水提物组、低浓度顺铂组、空白对照组,用CCK-8法检测药物作用48 h后对细胞存活率的影响,用RT-PCR检测细胞内Bcl-2、Bax、Survivin基因表达变化。结果:南方红豆杉水提物可增加顺铂对A549细胞的抑制作用,且随着药物浓度的增加,其抑制作用逐步增强;顺铂联和红豆杉通过下调Bcl-2,Survivin,并上调Bax的表达,而促进细胞发生凋亡,具有抗肿瘤作用。结论:南方红豆杉水提物联合顺铂能抑制A549细胞生长,其机制可能为凋亡相关基因Bcl-2,Survivin的降低,促凋亡基因Bax的增高有关。Objective： To study the inhibitory effect of aqueous extract of Taxus chinensis. var combining Cisplatin（ DDP） on vitro cultured human lung carcinoma A549 cell and to explore its mechanism.Methods： The A549 cells were divided into different concentrations of DDP groups,different concentrations of aqueous extract of Taxus chinensis.var groups and blank group,detecting drug effect of 48 h with the method of CCK-8 and the effect on cell survival. Based on the above results,A549 cells were divided into low concentration of DDP combining different concentrations of aqueous extract of Taxus chinensis.var groups,low concentration of DDP group,blank control group,detecting drug effect of 48 h with the method of CCK-8 and the effect on cells survival.The gene expressions of Bcl-2,Bax and Survivin were examined by RT-PCR.Results： Aqueous extract of Taxus chinensis. var combining DDP inhibited the proliferation of A549 cells and with the increase of drug concentration,the inhibitory effect gradually increased. Aqueous extract of Taxus chinensis.var combining DDP had antitumor effect by down regulating the expressions of Bcl-2,Survivin and up-regulating the expression of Bax,and then promoting cells apoptosis.Conclusion： Aqueous extract of Taxus chinensis.var combining DDP can inhibit the growth of A549 cells. The mechanism may be to decrease the apoptosis related gene Bcl- 2,Survivin and to increase the apoptosis gene Bax.
