摘  要:目的:研究淫羊藿对大鼠骨量及骨髓基质干细胞增殖、分化过程中Cbfa1和Osterix表达的影响。方法:24只雌性SD大鼠被随机分成观察组和对照组各12只,其中观察组给予淫羊藿0.5 g/0.1 kg,每天1次灌胃,持续两周;对照组给予相同剂量的生理盐水。于分组时间将大鼠处死,取右侧股骨远侧段用于骨组织形态计量学检测;抽取左侧股骨骨髓用于细胞培养,于培养21 d时提取细胞总RNA,经PCR方法分离获得cbfa1,osterix扩增产物。结果两组大鼠的骨形态计量学进行比较,观察结果表明,观察组大鼠骨组织计量学动态参数以及静态参数BV/TV、BFR/BS、tb.th、MAR比着对照组明显要高,差异具有统计学意义(P<0.05)。两组大鼠的Cbfa1,Cbfb和OSX m RNA表达的差异比较,研究组Cb fa1 m RNA,OSX m RNA在各时间点的表达均明显增加(P<0.05),并且这种差异随SIM作用时间增加而增强,研究组与对照组Cbfbm RNA差异无统计学意义(P>0.05)两组大鼠的骨形态蛋白-2以及转化因子β1相比,骨形态蛋白-2观察组明显高于对照组,差异存在统计学意义(P<0.05),转化因子β1比较观察组明显高于对照组,差异存在统计学意义(P<0.05)。结论:骨组织形态计量学方法显示淫羊藿组促进骨量增加;细胞分子生物学显示淫羊藿能够促进成骨细胞分化增殖作用,为中药抗骨质疏松提供组织细胞和分子生物学基础证据。Objective: To study the effect of ZH on the proliferation and differentiation of bone marrow mesenchymal stem cells and bone marrow mesenchymal stem cells in rats by Osterix. Methods: 24 female SD rats were randomly divided into observation group and control group with 12 rats in each,of which the observation group given Epimedium 0.5 g / 0.1kg,every day a irrigation stomach and lasts for two weeks. The control group received the same dose of saline. In packet time will rats were from right femoral distal segments used for bone histomorphometry examination; collect the left femoral bone marrow for cell culture and in cultured for 21 days in total RNA was extracted from the cells and by polymerase chain reaction( PCR) isolated Cbfa1,osterix amplification products. Results: Two groups of rats bone morphology metrology comparison,observation results show that the observation group rats bone metrology dynamic parameter and static parameter BV / TV,eliminated BFRS / BS,TB,th,MAR significantly better than the control group,difference has statistical significance( P〈0.05). Two groups of rats Cbfa1,Cbfb and OSX m RNA expression difference comparison,the team Cb fa1 m RNA,OSX m RNA expression in various time points were significantly increased( P〈0.05),and these differences enhanced with the increase of SIM effect time,the team with the control group there was no statistical significance( P〉0.05) difference Cbfbm RNA protein- 2 two groups of rats bone form as well as the conversion factor beta 1,compared to bone protein- 2 form observation group is significantly higher than control group,the difference is statistically significant( P〈0.05),the conversion factor beta 1 comparative observation group is significantly higher than control group,the difference is statistically significant( P〈0. 05). Conclusion: Bone histomorphometric method shows that Epimedium group promoted increased bone mass; molecular biology shows that Epimedium can promote osteoblast proliferation differentiation
