摘  要:目的:观察同仁再造丸抗脑缺血的效应机制,为临床合理应用提供基础研究数据。方法:采用小鼠断头急性脑缺血实验和小鼠双侧颈总动脉结扎法,观察再造丸对小鼠急性脑缺血的影响。采用大鼠线栓法、大脑中动脉阻塞(MCAO)脑缺血模型,观察再造丸对大鼠大脑梗死面积、神经功能评分以及MAD、SOD和NO的影响。采用"胶原蛋白-Adr"诱发小鼠体内血栓形成模型观察再造丸对偏瘫形成率和死亡率的影响;采用急性血瘀模型大鼠观察对血液流变学及血小板聚集率的影响,评价药物的活血化瘀作用。结果:再造丸高、中、低剂量组均可增加小鼠断头后急性脑缺血的呼吸次数与呼吸持续时间;再造丸高、中剂量组可延长双侧颈总动脉结扎法小鼠急性脑缺血的存活时间。再造丸可显著减小MCAO大鼠梗死面积;降低缺血后4 h、24 h模型大鼠神经功能行为学评分;各给药组均可显著降低模型大鼠血清MDA含量;再造丸高剂量组可提高MCAO模型大鼠血清SOD活力;再造丸低剂量组可显著升高大鼠血清NO水平。再造丸可减少尾静脉注射"胶原蛋白-Adr"形成体内血栓所致小鼠5 min内的死亡率,提高15 min内的偏瘫恢复率。再造丸可降低血瘀模型大鼠全血血流变切变率,降低ADP诱导的血小板聚集率。结论:再造丸对脑缺血有保护作用,保护机制可能与抗氧化及改善血液流变学相关。Objective: To observe the effects of Tongrentang Zaizaowan on the pharmacodynamic. Methods:The decapitation ischemia model and bilateral carotid artery ligation was used to observe the effects of Tongrentang Zaizaowan on acute cerebral ischemia. MCAO model was induced by a middle cerebral artery occlusion, infract area, ethological index, MDA, SOD and NO in serum were observed. The number of paralysis or dead was calculated in mice, induced by tail intravenous injection of a mixture of collagen and Epinephrine.The effect of activating blood circulation to dissipate blood stasis was observed by haemorheology and blood clotting. Results: Zaizaowan significantly raised breathing time and frequency; Zaizao high dose group can significantly prolong the survival time of bilateral carotid artery ligation in mice with acute cerebral ischemia.Zaizaowan remarkably down-regulated the infarct size, and ethological index after ischemia 4 h and 24 h, the level of MDA in serum, up- regulated the level of SOD and NO on MCAO rats. Zaizao can reduce the mortality within 5 min, and increase the recovery rate within 15 min induced by tail vein injection "collagenAdr" thrombus formation. Zaizaowan can decrease the blood viscidity of the rats, reduce the Platelet Aggregation rate induced by ADP. Conclusions: Zaizaowan has protective effect on cerebral ischemia, and its mechanisms may be related to anti-oxidant and improving hemorrheology capacity.
