摘 要:目的:比较川芎-天麻药对不同提取物对血瘀证偏头痛大鼠的药效学差异。方法:雄性SD大鼠随机分为9组,即为正常对照组、模型对照组、阳性药盐酸氟桂利嗪(西比灵)组、川芎-天麻(4∶1,)水提物组、川芎-天麻(1∶1)水提物组、川芎-天麻(1∶4)水提物组、川芎-天麻(4∶1)醇提物组、川芎-天麻(1∶1)醇提物组、川芎-天麻(1∶4)醇提物组。大鼠冰水浴后皮下注射盐酸肾上腺素(0.8 mg/kg)和硝酸甘油(10 mg/kg)复制血瘀证偏头痛模型,比色法测定大鼠血清一氧化氮合酶(NOS)活力与一氧化氮(NO)水平,Elisa法测定血清降钙素基因相关肽(CGRP)、内皮素(ET)、血栓素B2(TXB2)、6-酮-前列腺素F1α(6-keto-PGF1α)水平并计算NO/ET。结果:模型组血清CGRP水平、NOS活力、NO、TXB2含量及NO/ET比值升高,6-keto-PGF1α含量降低,与正常组比较差异有显著性(P<0.05);各比例提取物组间比较,各比例组醇提物NO水平及NO/ET比值均低于同比例水提物(P<0.05),6-Keto-PGF1α(4∶1组)水平高于同比例水提物(P<0.05)。结论:川芎-天麻不同提取物对血瘀型偏头痛均有一定疗效,不同提取物组间疗效有差异,综合多项指标考虑,醇提物疗效优于水提物。Objective:To study the pharmacodynamic differences of migraine rat models with blood stasis treated with different proportion of Rhizoma Ligusticum chuanxiong and Gastrodia Elata(4∶1,1∶1,1 ∶ 4)extracted with water and 70% alcohol. Methods:SD rats were randomly divided into 9 groups:normal group,model group,Flunarizine group and different propotion of Rhizoma Ligusticum chuanxiong and Gastrodia Elata(4∶1),(1∶1),(1∶4)water and alcohol extract group. Adrenaline(s.c. 0.8 mg/kg)and ice water bath were used to establish acute blood stasis model,migraine models were copied by glyceryltrinitrate(s.c. 10 mg/kg). Then determined the content of CGRP,NOS,NO,ET,TXB2,6-ketoPGF1α in rat serum. Results:In model group,The levels of CGRP,NOS,NO,TXB2 and the ratio of NO/ET raised(P0.05)and the level of 6-keto-PGF1α reduced(P0.05). The level of NO and the ratio of NO/ET were higher in water extraction than alcohol extraction(P0.05),whereas the level of 6-ketoPGF1α was(4∶1)lower than alcohol extraction(P0.05). Conclusion:Each of the medicated groups was effective on the model of migraine with blood stasis and alcohol extractions were more effective than water extractions.
