摘  要:目的:将淫羊藿各有效成分进行提取分离,拆分成互不交叉的各段成分,并对各段成分进行体内抗衰老、体外抗氧化活性的筛选。方法:将淫羊藿生药材通过聚酰胺树脂进行分离,运用HPLC方法验证各段成分交叉情况,通过构建D-半乳糖所致的衰老小鼠动物模型,来考察淫羊藿各段成分对衰老小鼠血、肝、脑中SOD、MDA和GSH-px的变化;并且观察不同浓度的淫羊藿各段成分对体外DPPH自由基的清除作用。结果:淫羊藿全成分给药组对衰老模型小鼠的血、肝中MDA含量显著降低,对其中的GSH-px酶活力有显著的升高,对血、肝、脑中的SOD酶活力有显著的升高(P<0.05);淫羊藿30醇段给药组和95醇段给药组对衰老模型小鼠的肝组织中SOD酶活力水平有明显升高(P<0.05),其中30醇段给药组可使衰老小鼠肝中MDA含量非常显著降低(P<0.01),95醇段给药组可使衰老小鼠血中MDA含量显著降低(P<0.05);此外淫羊藿水段给药组仅在肝中GSH-px酶活力水平上有显著升高(P<0.05),因此各组成分抗衰老作用大小依次为:全成分组>30醇组>95醇组>水段组>多糖组。体外DPPH自由基清除实验中发现,随浓度增加,淫羊藿各段成分对DPPH自由基清除作用也随之增大,其中全成分组分对DPPH自由基清除作用最显著,当浓度达到5 g/m L时,清除率达到89.46%,各段清除作用大小依次为:全成分组>水段组>30醇组>95醇组>多糖组。结论:淫羊藿可分离成互不交叉的五组化学成分,分别为淫羊藿全成分组、水段组、30醇段组、95醇段组和多糖组,其中全成分组分在抗衰老和抗氧化两方面都作用最强,其他各段成分中30醇段组分抗衰老活性最佳,水段组分抗氧化活性最好。Objective:Various of effective constituent of epimedium were extracted and separated.Each section of uncrossed ingredients was filtrated for active of anti-aging in vivo and active of antioxidation in vitro. Method:Crude epimedium was separated by amilan,and the cross situation of every section was verified by HPLC. By means of building animal model of aging mice induced by d-galactose,the influence were evaluated of every section of epimedium on SOD,MDA and GSH-px in blood,liver and brain of mice.The scavenging action were observed for DPPH of different concentration of each section. Result:In entire ingredient section,the MDA in blood and liver decreased significantly,and the activity of GSH-px enzyme increased significantly.The activity of SOD enzyme in blood,liver and brain increased significantly(P0.05);in 30% and 95% alcohol section,the level of SOD enzyme in liver increases significantly(P0.05),and 30% acohol section reduced MDA in liver very significantly(P0.01),95% section reduced MDA in blood significantly(P0.05).In addition,water section selectly rose the level of GSH-px enzyme in liver significantly(P0.05). So,the sequence of anti-aging activity is:entire ingredient30% alcohol95% alc oholwaterpolysaccharide.From the DPPH scavenging experiment in vitro,the author found that the effect of every group on scavenging DPPH increased with the concentration increased,and the entire ingredient was most significantly between groups.The sequence was:entire ingredientwater30% alcohol95% alcoholpolysaccharide. Conclusion:Epimedium was extracted to five sections:entire ingredient,water,30% alcohol,95% alcohol and polysaccharide. The entire ingredient manifested most strong effect on antiaging and antioxidation,and the 30% alcohol group showed excellent anti-aging between the other groups.The water group displayed good antioxidation.
