泽泻汤对血管平滑肌细胞迁移及相关因子MMP-2和MMP-9表达的影响

摘　　要：目的:观察泽泻汤对大鼠血管平滑肌细胞(VSMCs)的迁移作用及其相关因子MMP-2、MM-9表达的影响。方法:通过体外ox-LDL(50mg/L)诱导VSMCs,建立VSMCs迁移的动脉粥样硬化(AS)细胞模型,采用20%泽泻汤含药血清干预后,采用划痕实验测定VSMCs迁移,采用Western blot检测MMP-2、MMP-9蛋白表达情况。结果:VSMCs在ox-LDL的刺激下,其迁移能力与对照组比较有明显提升;加入泽泻汤含药血清后,细胞迁移距离变短,其迁移能力受到抑制。ox-LDL组MMP-2、MMP-9蛋白表达较对照组明显增强,而泽泻汤血清干预后MMP-2、MMP-9蛋白表达较ox-LDL组则明显受到抑制。结论:泽泻汤可抑制ox-LDL诱导的VSMCs迁移和MMP-2、MMP-9蛋白表达,提示泽泻汤抑制VSMCs迁移的作用机制可能与影响MMP-2和MMP-9的表达有关。Objective：To observe the effect of Alisma Decoction on the migration of vascular smooth muscle cells（VSMCs）,and on the expressions of its correlation factors-matrix metalloproteinase（MMP）-2and MMP-9.Methods：The model of Atherosclerosis VSMCs migration was established after induction with oxidized low density liprotein（ox-LDL）（50mg/L）and then the cells were intervened with 20% Alisma Decoction-containing serum.Cell migration was studied through a scratch-wound assay.The matrix metalloproteinase-2and matrix metalloproteinase-9activities were analyzed by Western blot.Results：Compared with the control group,the migration of VSMCSs was increased in the ox-LDL group.However,the migration of VSMCS was decreased after treated with the Alisma Decoction-containing serum compared with the ox-LDL group.The expression of MMP-2and MMP-9in the ox-LDL group was much more greatly than the control group and inhibited significantly after treated with the Alisma Decoction-containing serum.Conclusion：Alisma Decoction can inhibit the migration of VSMCS induced by ox-LDL and the expression of MMP-2and MMP-9.The results indicate that the inhibitory effect of Alisma Decoction on VSMCS migration may be related to its influence on the cytokine expression and regulation to the metabolism of extracellular matrix.
