摘  要:【目的】解析鹅圆环病毒(Goose Circovirus,Go CV)广西流行毒株的全基因组序列及其遗传进化地位,掌握广西Go CV的流行情况,为今后更有效防控Go CV提供理论依据。【方法】分3段对阳性组织样品中的Go CV全基因组进行PCR扩增、克隆和测序,并对拼接得到的Go CV广西流行毒株全基因组进行核苷酸组成、基因组结构及遗传变异分析。【结果】Go CV广西流行毒株(GXTD254)全基因组长度为1822 nt,编码4个主要开放阅读框:ORF V1(Rep蛋白,293个氨基酸)、ORF V2(37个氨基酸)、ORF C1(Cap蛋白,250个氨基酸)和ORF C2(99个氨基酸),且Rep蛋白较Cap蛋白相对保守。遗传进化分析结果表明,GXTD254毒株与4株浙江永康分离毒株(ZJyk1、ZJyk2、ZJyk3和ZJyk4)、1株浙江象山分离毒株(ZJxs1)的遗传距离最接近,同属于一个遗传分支,但与我国台湾的大多数分离毒株(TW1021024G、TWGB21-9、TW6、TW9、TW8和TW1020111GB)及另外2株浙江象山分离毒株(ZJxs3和ZJxs5)的遗传距离较远。【结论】目前我国流行的Go CV存在多个分支,广西流行毒株与部分浙江分离毒株在亲缘关系上比较接近。【Objective】The genome sequence and genetic evolutionary status of goose Circovirus(Go CV) in Guangxi were investigated to understand epidemic situation, in order to provide theoretical basis for effective prevention and control of Go CV. 【Method】The 3 fragments were amplified using PCR technique, and sequenced. Then 3 fragments were connected together to obtain full-length genome of epidemic strain. And the nucleotide composition, genomic structure and heritable variation of full-length genome were analyzed. 【Result】The results showed that, the genome of Go CV(Guangxi epidemic strain GXTD254) was 1822 nt in length, which coded four major open reading frames: ORF V1(Rep protein, 293 amino acids), ORF V2(37 amino acids), ORF C1(Cap protein, 250 amino acids), ORF C2(99 amino acids), and the sequences of Rep and Cap proteins were relatively conservative. In addition, the genetic evolution analysis showed that GXTD254 was clustered into a same branch with 4 strains(ZJyk1, ZJyk2, ZJyk3, ZJyk4) isolated from Yongkang, Zhejiang and 1 strain(ZJxs1) isolated from Xiangshan, Zhejiang, with the closest genetic relationship. But it had greater genetic distance with most of strains(TW1021024G, TWGB21-9, TW6, TW9, TW8 and TW1020111GB) isolated from Taiwan and 2 strains(ZJxs3 and ZJxs5) isolated from Zhejiang province. 【Conclusion】At present, the epidemic Go CV strains can be clustered into several branches, which have the closer genetic relationship with Go CV strains isolated from Zhejiang province.
