摘　　要：目的探讨创伤后应激障碍(PTSD)大鼠前额叶皮质Npas4的表达变化情况。方法采用单程长时应激(SPS)制备PTSD大鼠模型,将PTSD大鼠分为4组:SPS1d、SPS3d、SPS7d和SPS14d组,正常饲养大鼠作为对照组;利用免疫荧光染色、Western Blot和real-time PCR检测Npas4蛋白及mRNA表达情况。结果在SPS后第1d、3d和7d,PTSD大鼠前额叶皮质Npas4蛋白和mRNA表达水平显著低于正常对照组,SPS14d时恢复到正常对照组水平。结论 Npas4低表达可能与PTSD发病机制相关。Objective To investigate the expression changes of Npas4 in prefrontal cortex of post-traumatic stress disorder（PTSD） rats.Methods The PTSD models were established by single prolonged stress paradigm（SPS）,divided into four groups：SPSld,SPS3 d,SPS7d and SPS14 d groups,normal fed rats as a control group.The expressions of Npas4 protein and mRNA in prefrontal cortex were detected by immunocytochemistry,Western blot and real-time PCR.Results The expression levels of Npas4 protein and mRNA in prefrontal cortex were significantly decreased in SPS1 d,SPS3d and SPS7 d rats than in normal control rats,but upregulated in SPS14 d rats near to normal control rats.Conclusion The downexpression of Npas4 in prefrontal cortex might be related to the pathogenesis of PTSD.
