摘  要:目的:研究尼古丁戒断期运动干预对大鼠抑制能力和大脑α7 烟碱乙酰胆碱受体(nAChRs)蛋白表达的影响,以探讨抑制能力与尼古丁CPP依赖性和nAChRs蛋白表达之间的关系。方法:30只4周龄雄性SD大鼠随机分为安静组和中等强度运动组。采用条件位置偏爱(CPP)建立大鼠尼古丁依赖模型,Go/NoGo范式评价戒断大鼠的抑制能力,Western blotting蛋白免疫印迹法检测前额叶皮层和海马组织α7 nAChRs蛋白表达水平。结果:1)构建了大鼠Go/NoGo范式和尼古丁CPP依赖性模型。2)与安静组相比,运动组大鼠CPP得分(s)显著降低(156.27±12.07vs 83.6±14.85,P<0.01),Go任务正确率(%)没有变化,但NoGo任务正确率(%)明显提高(42.67±3.53vs 63.43±3.92,P<0.01)。3)安静组和运动组大鼠海马组织a7 nAChRs表达水平没有发生明显变化,而运动组大鼠前额叶皮层α7 nAChRs蛋白表达显著上调(0.7±0.06vs 1.23±0.1,P<0.01)。4)尼古丁戒断大鼠的抑制能力与CPP偏好值负相关(R2=0.64,P<0.01),与前额叶皮层α7 nAChRs蛋白表达水平正相关(R2=0.44,P<0.05)。结论:连续10天的中等强度跑台运动可能通过特异性上调前额叶皮层a7 nAChRs蛋白表达,增强尼古丁戒断大鼠的行为抑制能力,进而降低尼古丁依赖性。Objective:To explore the effect of exercise on inhibition ability and the expression of a7 nicotinic acetylcholine receptors (nAChRs) during nicotine abstinence in rats, and discuss the relevant relationships between them. Methods: 30 four-week-old male Sprague-Dawley rats were randomly divided into sedentary group and moderate intensity exercise group. It was per- formed with conditioned place preference (CPP) to establish nicotine dependence model in rats. The study applied Go/NoGo task to evaluate inhibition ability during nicotine abstinence periods, and the expression of a7 nAChRs in prefrontal cortex and hippocampus were detected by Western blotting. Results: 1 ) Go/NoGo paradigm and nicotine-induced CPP were estab- lished successfully. 2) Compared to the sedentary group, the preferred values of rat nicotine-CPP (secs) from exercise group was significantly reduced (156.27±12.07 vs 83.6±14.85, P〈0.01) ;The accuracy ratio of Go task (%) did not change, while the accuracy ratio of No- Go task (%) increased observably (42.67+_3.53 vs 63.43!3.92,P〈0.01). 3) The expression of a7 nAChRs in both groups did not change in the hippocampus, but it was significantly up-regulated in the prefrontal cortex in exercise group (0. 7±0.06 vs 1.23±0. 1, P〈0.01). 4) The inhibition ability of rats during nicotine abstinence periods was negatively corre-lated with the preferred values of CPP (R2 = 0.64, P〈0.01), and positively correlated with the prefrontal cortex a 7 nAChRs expression (R2 = 0.4 4, P〈 0.0 5). Conclusion :10-day mod- erate intensity treadmill running can promote rats inhibition ability during nicotine abstinence period, and reduce dependence of nicotine. These effects of exercise may be due to the up-regu-lation of a7 nAChRs.
