摘  要:目的研究大鼠胚胎着床后短暂抑制雌激素合成对分娩结局的影响。方法大鼠胚胎着床后GD 6.5-8.5给予雌激素抑制剂来曲唑灌胃(实验组)或溶媒灌胃(对照组),观察其对胎儿发育和分娩结局的影响。结果实验组和对照组的窝仔数和胎(仔)鼠重量比较无统计学差异(P〉0.05);实验组孕鼠在GD 22.5均不能正常分娩,在GD 23.5体力衰竭死亡,而于GD 22.5时剖腹取出的仔鼠体重和发育正常,且可代乳存活;GD 21.5和GD 22.5时,对照组外周血孕酮水平快速下降至(6.7±4.4)nmol/L和(5.6±3.5)nmol/L,而实验组孕酮水平分别为(19.7±7.0)nmol/L和(18.0±4.2)nmol/L,显著高于对照组(P〈0.05);两组孕鼠均分别在GD 19.5、GD 20.5以及GD 20.5、GD 21.5连续2d给予米非司酮,均分别于GD20.5和GD 21.5启动分娩并正常生产。结论大鼠胚胎着床后给予来曲唑短暂抑制雌激素合成可导致孕鼠难产,妊娠末期给予米非司酮可以挽救难产结局。Objective: To determine the effect of transient inhibition of estrogen production in post-implantation period on pregnancy outcome in normal rats.Methods: Letrozon,an aromatase inhibitor,was intragastrically administered to pregnant rats at a dose of 0.16 mg/kg/day from gestation day(GD)6.5to 8.5(treated group),or vehicle as a control(control group).Fetal development and pregnancy outcomes were examined.Results: There was no significant difference in litter size and fetal weight between letrozole treated group and control group(P〉0.05).The pregnant rat in control group normally completed parturition at GD 22.5,but the rats in letrozole treated group cannot deliver normally at GD 22.5,and ended in death with physical exhaustion at GD 23.5.When cesarean was performed at GD 22.5in treated rats,it was found that offspring weight and development were normal,and the pups can be survived from milk replacer.The serum progesterone levels of maternal rats in control group rapidly declined to(6.7±4.4)nmol/L and(5.6±3.5)nmol/L at GD 21.5and GD 22.5respectively,but the progesterone levels in treated group was(19.7±7.0)nmol/L and(18.0±4.2)nmol/L respectively.There was a significant difference between the treated and control group(P〈0.05).When mifepristone of 10 mg/kg/day was subcutaneously injected for two days at GD 19.5 & GD 20.5 and GD 20.5 & GD 21.5 respectively,all maternal rats in the treated and control group completed parturition at GD 20.5 and GD 21.5 respectively.Conclusions: These results demonstrated that transient inhibition of estrogen synthesis in postimplantation period in maternal rats can result in dystocia at term.And administered mifepristone at term can rescue the dystocia.
