摘 要:目的观察厚朴冷浸液对白色念珠菌(Candida albicans)黏附性及生物膜形成的影响,以探讨其能否用于治疗白色念珠菌相关龋病。方法运用微孔板法复制白色念珠菌体外黏附及生物膜模型,采用XTT减低法评价不同浓度厚朴冷浸液对白色念珠菌黏附、生物膜形成的影响;光镜下观察生物膜内白色念珠菌形态学变化。结果厚朴冷浸液对白色念珠菌的黏附具有抑制作用,在同一时间点其抑制作用具有浓度依赖性,但是随着时间延长,各组药物对白色念珠菌的黏附作用均减弱;厚朴冷浸液对白色念珠菌形成的生物膜具有抑制作用,其作用具有浓度依赖性,除7.80 mg/L浓度组外,其余各组与阴性对照组相比均有统计学意义(P<0.05),125.00 mg/L浓度组对生物膜的抑制率达到60.27%;厚朴冷浸液使生物膜内白色念珠菌脱落,芽管形成受抑制。结论厚朴冷浸液对白色念珠菌早期黏附和中期生物膜形成有抑制作用。Objective To observe the impact of Magnoliae cortex quenching liquidon Candida albicans adhesion and biofilm formation in order to assess its anti-caries effect. Methods The 96-well microtiter plates were used to produce C. albicans in vitro adhesion and biofilm model,XTT reduction assay was applied to evaluate the effect of the different concentrations of Magnoliae cortex quenching liquid on C. albicans adhesion and biofilm formation. Light microscope was employed to observe the morphological changes of Candida albicans in biofilm.Results Magnoliae cortex quenching liquid had inhibitory effect on C. albicans adhesion,its action was concentration-dependent at the same time point. However,with the time prolonged,the drug for C. albicans adhesion effect weakened. Magnoliae cortex quenching liquid had inhibitory effect on C. albicans biofilm formation; its action was concentration dependent. Compared with negative control group had statistical significance( P ﹤0. 05) in addition to the concentration of 7. 8 mg / l group. Concentration of 125. 00 mg / l group of inhibition rate reached 60. 27 %. Magnoliae cortex quenching liquid led to C. albicansin the biofilm shedding and inhibited germ tubes formation. Conclusion Magnoliae cortex quenching liquid has inhibitory effect on C. albicans early adhesion and middle of biofilm formation.
