寻常型银屑病湿热证与血热证的差异血清蛋白质组学表达初探

摘　　要：目的:应用同位素相对标记与绝对定量技术(iTRAQ)技术对寻常型银屑病(血热证、湿热证)患者与健康人差异且共有蛋白进行分析,探求两证之间差异性表达蛋白,从蛋白质组学角度揭示两证之间差异。方法:取寻常型银屑病血热证、湿热证患者血清,予纯化等处理后,SDS-PAGE电泳分离,经i TRAQ特异性标记酶解后的肽段,采用串联质谱分析与健康人比较寻找差异且两证共有蛋白,并将这些蛋白进行组间比较。结果:发现了19个显著性差异蛋白(P<0.05),其中,强显著性差异的蛋白有6个(P<0.01)。结论:寻常型银屑病血热证与湿热证血清中存在表达差异性蛋白质,如糖蛋白类、脂蛋白类、免疫类等。是否为证型之间的特征性差异蛋白,有待进一步证实。Objective：To analyze the differential and identical proteins in psoriasis vulgaris patients with blood heat syndrome and damp-heat syndrome and healthy people by using iTRAQ technology,in order to analyze the differentially expressed proteins between the two syndromes,and reveal the differences between the two syndromes from the perspective of proteomics.Methods：The serum samples were obtained from psoriasis vulgaris patients with blood heat syndrome and dampheat syndrome.After purifying and separating of SDS-PAGE electrophoresis,the peptide was signed with iTRAQ,and differential and identical proteins in psoriasis vulgaris patients with blood heat syndrome and damp-heat syndrome and healthy people were analyzed by using tandem mass spectrometry.Results：Ninteen differential proteins were identified（P〈0.05）,in which there were6 differential proteins with significant differences（P〈0.01）.Conclusion：There are some differential proteins such as antibacterial peptide protein group,CAM protide,VEGF receptors and TGF-β1 in serum of psoriasis vulgaris patients with blood heat syndrome and damp-heat syndrome.Whether those proteins were the characteristic differential proteins of the two syndromes,it remains to be further study.
