金欣口服液对呼吸道合胞病毒感染大鼠JAK/STAT信号通路P-STAT1、P-STAT2调控的研究

摘　　要：目的:研究金欣口服液对RSV感染大鼠体内Ⅰ型干扰素相关蛋白STAT1、STAT2和P-STAT1、P-STAT2表达的调控趋势。方法:将大鼠分为正常对照组(A组)、感染模型组(B组)、金欣口服液低剂量组(C组)、金欣口服液中剂量组(D组)、金欣口服液高剂量组(E组)、预防组(F组)、利巴韦林组(G组)7组,成功造模后,分别采用ELISA和RT-PCR检测各组大鼠肺组织内P-STAT1、P-STAT2和STAT1、STAT2的表达。结果:感染模型组较正常对照组、预防组和金欣口服液低剂量组未能上调大鼠体内P-STAT1和P-STAT2蛋白表达;而金欣口服液中、高剂量及利巴韦林组明显上调感染模型组RSV大鼠体内的P-STAT1和P-STAT2蛋白的表达(P<0.05,P<0.01)。结论:实验表明金欣口服液可通过上调P-STAT1和P-STAT2的表达,诱导IFN的产生,发挥抗病毒作用。Objective：To study the regulatory effect of Jinxin Oral Liquid on type I interferon related proteins such as STAT1,STAT2,P-STAT1 and P-STAT2 in RSV infected rats.Methods：rats were randomly divided into control group,model group,low,middle and high dosages of Jinxing Oral Liquid groups,prevention group and ribavirin group.After establishing the rat models,Elisa and RT-PCR were used to test the expression of P-STAT1,P-STAT2,STAT1 and STAT2 in lung tissue of rats in all the groups.Results：The expression of P-STAT1 and P-STAT2 of rats in low dosage of Jinxing Oral Liquid groups and prevention group was not increased,and the expression of P-STAT1 and P-STAT2 of rats in middle and high dosages of Jinxing Oral Liquid groups and ribavirin group was increased significantly（P〈0.01）.Conclusion：The results show that Jinxing Oral Liquid could induce the occurrence of IFN and play the role of antiviral by up-requlatinq the expression of P-STAT1 and P-STAT2.
