摘  要:目的研究磷酸肌酸不同给药方式对于糖尿病心肌缺血再灌注大鼠内皮素(ET-1)和一氧化氮(NO)含量的影响。方法建立糖尿病病程2周鼠和8周鼠模型各60只,两组大鼠分别随机分为四组(n=8):假手术组(S组);I/R+生理盐水组(I/R组);I/R+Pcr单次给药组(单次给药组);I/R+Pcr多次给药组(多次给药组)。S组:只在左冠脉部位穿线而不结扎;I/R组:结扎前30 min腹腔注射生理盐水;单次给药组:结扎前30 min,按2 g/kg一次性腹腔注射Pcr;多次给药组:按1 g/kg每日一次腹腔注射Pcr,连续5 d,注药30 min后结扎冠脉;各组均缺血30 min,再灌注2 h。再灌注结束后,分别测定大鼠血浆中ET-1和NO的含量。结果同时间内,与S组相比,I/R组中ET-1的含量均增加(P<0.05),NO的含量均减少(P<0.01);与I/R组相比,单次给药组中ET-1含量减少(P<0.05),单次给药组和多次给药组中NO含量均增加(P<0.01);与多次给药组相比,单次给药组ET-1含量均减少(P<0.05),NO含量均增加(P<0.01)。8周鼠S组和I/R组分别与2周鼠对应组比较,ET-1含量均增加(P<0.05),NO含量均明显减少(P<0.01)。结论磷酸肌酸可减少糖尿病大鼠I/R后血浆ET-1的含量,增加NO的含量,单次大剂量给药效果优于多次小剂量给药。Objective To investigate the effect of different administration ways of phosphocreatine( Pcr) on endothelin-1 and NO in diabetic rats with myocardial ischemia-reperfusion injury( I / R). Methods The rats were fed with high lipid to establish the diabetic models: 2-week disease course and 8-week disease course. The two kinds of rats were respectively randomized into four groups( n = 8) :sham group,I / R group,single administration of Pcr group,and multiple administration of Pcr group. The artery of rats was not ligated in sham group. The rats in I / R group were given saline 30 min before ligation. The rats were administered with 2 g / kg Pcr once in single Pcr group. The rats were given 1 g / kg Pcr once a day for 5 d in multiple Pcr group. The coronary artery was ligated to induce the myocardial ischemia reperfusion in all rats. The artery was occluded for 30 min by tightening the ligature and followed by reperfusion for2 h. ET-1 and NO were determined after the reperfusion. Results Compared with sham group,the serum level of ET-1 was significantly increased at week 2 and 8 in I / R group( P〈0.05),while the content of NO was significantly decreased( P〈0.01). Compared with I /R group,the serum level of ET-1 in single Pcr group was decreased( P〈0.05),while the contents of NO in single Pcr group and multiple Pcr group were significantly increased( P〈0.01). Compared with multiple Pcr group,ET-1 was decreased in single Pcr group( P〈0.05),while NO was significantly increased( P〈0.01). In sham group and I/R group,the contents of ET-1 were higher in model rats at 8 week than those at 2 week( P〈0.05),while the contents of NO were significantly lower( P〈0.01). Conclusion Phosphocreatine could reduce the serum level of ET-1 and increase the content of NO in diabetic rats,which may be one of the mechanisms of myocardial protection. And the effect of single administration and high dose of Pcr is better than that of multiple administration and small dose.
