摘 要:目的探讨人参皂苷Rg1对大鼠离体心脏缺血再灌注损伤的减轻作用及机制。方法制备大鼠离体心脏I/R模型,将18只大鼠随机分为正常对照组、I/R组、人参皂苷Rg1组。100μmol/L人参皂苷Rg1进行预处理10 min,观察血流动力学指标左心室内压最大上升/下降速率和心率压力乘积的变化,检测灌流液中肌酸激酶(CK)、乳酸脱氢酶(LDH)活性。Western blotting检测心肌组织PI3K下游激酶Akt、ERK1/2及其磷酸化水平的变化,磷酸化GSK-3β的蛋白表达。结果与I/R组比较,人参皂苷Rg1预处理显著改善心脏功能,降低灌流液中CK、LDH活性(P<0.05);升高p-Akt和p-GSK-3β的蛋白表达(P<0.05)。结论人参皂苷Rg1对大鼠离体心脏缺血再灌注损伤有减轻作用,其机制与激活PI3K-Akt通路有关。Objective To investigate cardioprotection effects of Ginsenoside Rgl on ischemia/reperfusion (I/R) injury in isolated hearts of rats, and to explore its action mechanism. Methods The Langendorff models of myocardial I/R injury in rats were established. The 18 rats were randomly divided into three groups: control group, I/R group and Ginsenoside Rgl group, with 6 rats in each group. The rats' hearts in Ginsenoside Rgl group were pretreated with Ginsenoside Rgl 1001~mol/L for 10rain. The cardiac indexes including maximal rise/fall of left ventricular pressure ( ± dp/dtmax) and rate pressure product (RPP) were measured,and the activities of creatine kinase (CK) and lactate dehydrogenase (LDH) were detected. The changes of phosphorylation of protein kinase B/Akt, and its downstream target glycogen synthase kinase 313 (GSK-313) and extracellular signal-regulated kinasel/2 (ERK1/2) were detected after 10 -minute reperfusion. Results As compared with those in I/R group, the activities of CK, LDH in Ginsenoside Rgl group were significantly decreased and cardiac function was obviously improved ( P 〈 0. 05 ), moreover, the expression levels of p-Akt and p-GSK-313were significantly increased ( P 〈 0.05). Conclusion Ginsenoside Rgl can alleviate I/R injury in isolated hearts of rats, and its action mechanism may be correlated to the activation of PI3K-Akt signaling pathway.
