摘 要:目的探讨黄芪注射液对1型糖尿病小鼠胰岛β细胞凋亡的保护作用。方法将32只小鼠按随机数字表法分为正常对照组、模型组、小剂量黄芪组、大剂量黄芪组各8只。除正常对照组外,其余各组小鼠腹腔注射STZ溶液制备糖尿病模型。注射后1周,小、大剂量黄芪组分别腹腔注射黄芪注射液30、60g/(kg·d),正常对照组和模型组小鼠腹腔注射等体积生理盐水,连续腹腔注射4周。采用ELISA法检测血清NO、iNOS及胰岛素水平;末端脱氧核苷酰基转移酶介导dUTP切口末端标记(TUNEL)法检测胰岛β细胞凋亡情况。结果与模型组比较,小、大剂量黄芪组小鼠血清iNOS[(21.38±4.48)μmol/L、(17.00±3.05)μmol/L比(26.62±2.48)μmol/L)]、NO[(25.81±2.09)μmol/L、(18.84±3.95)μmol/L比(30.34±2.53)μmol/L]水平降低(P〈0.05);小、大剂量黄芪组小鼠胰岛素水平[(312.19±66.91)μmol/L、(330.14±56.06)μmol/L比(273.56±43.54)μmol/L]增高、胰岛β细胞凋亡率[(23.00±16.58)%、(27.38±18.79)%比(31.25±18.19)%]减少,但差异无统计学意义(P〉0.05)。结论黄芪注射液可显著降低1型糖尿病小鼠iNOS活性,减少NO生成,但对胰岛β细胞凋亡的保护作用不明显。Objective To investigate the therapeutic effect ofastragalus injection on β cells apoptosis of type 1 diabetes mice. Methods 32 mice were randomly divided into a control group, a diabetes group, a small dose group and a large dose group of astraglus. Except the control group, mice in the other 3 groups received intrapedtoneal injection of STZ in order to induce diabetic mellitus. Then one week after injection of STZ, mice in the small dose group and the large dose group of astraglus were given 30, 60 g/(kg·d) doses of astragalus injection, while the other 2 groups were injected into an equal volume saline for 4 weeks. Serum NO, inducible nitric oxide synthase (iNOS) and insulin levels were detected by enzyme linked immunosorbent assay method, and apoptosis was measured by using a TUNEL assay. Results Compared with the diabetes group, the serum NO (25.81 ± 2.09 μmol/L, 18.84 ± 3.95 μmol/L vs. 30.34 ± 2.53 μmol/L) and iNOS (21.38 ±4.48 μmol/L, 17.00 ± 3.05 μmol/L vs. 26.62± 2.48 μmol/L) levels were significantly reduced in the small dose group and the large dose group of astragalus (P〈0.05); But the insulin levels (312.19 ± 66.91 pmol/L, 330.14 ± 56.06 pmol/L vs. 273.56 ± 43.54 pmol/L) and apoptosis of β cells (23.00% ± 16.58%, 27.38% ± 18.79% vs. 31.25% ± 18.19%) were not significant different among the experimental groups (P〉0.05). Conclusion Astragalus injection can reduce the iNOS activity and NO production, but can not effectively decreasing β cells apoptosis.
