摘 要:目的:探讨寿胎丸提高控制性超排卵(COH)小鼠子宫内膜容受性的作用机制。方法:采用促性腺激素释放激素激动剂+孕马血清促性腺激素+人绒毛膜促性腺激素建立COH小鼠模型,将其分为模型组、寿胎丸高、中、低剂量组、阿司匹林对照组,同时设正常对照组,在造模的同时,寿胎丸(高、中、低剂量)组分别按6、3、1g/kg寿胎丸灌胃;阿司匹林对照组按0.2g/kg阿司匹林灌胃;模型组、正常对照组以等体积生理盐水灌胃;每天1次,连用11天。采用HE染色、免疫组化法对子宫进行病理切片制作,通过Mias-2000图像分析系统检测子宫内膜Ang-1、Ang-2、Tie-2蛋白表达水平,并测量子宫内膜厚度及其血管数。结果:与正常对照组比较,模型组、寿胎丸高、中、低剂量组、阿司匹林对照组子宫内膜厚度、Ang-2表达无明显变化。与正常对照组比较,模型组子宫血管数、Ang-1表达、Tie-2表达明显减少(P<0.05);与模型组比较,寿胎丸高、中、低剂量组、阿司匹林对照组子宫血管数、子宫内膜Ang-1表达明显增多(P<0.05),寿胎丸高、中剂量组、阿司匹林对照组子宫内膜Tie-2明显增多(P<0.05)。结论:寿胎丸可能通过调节COH小鼠子宫内膜Ang及受体表达、增加子宫血管数量,进而提高COH小鼠子宫内膜容受性。Objective: To explore the mechanism of Shou Tai Wan in improving endometrial receptivity of mice with controlled ovarian hyperstimulation (COH). Method: COH model mice were established by using GnRH a + PMSG + hCG, and the model mice were divided into 6 groups, which included model group, high dose, medium dose or low dose Shou Tai Wan groups, aspirin control group and normal control group. As for Shou Tai Wan groups, the mice were gavage administrated Shou Tai Wan 6g, 3g, 1g/kg, respectively, once a day for 11 days. In aspirin group, mice were gavage administrated aspirin (0.2 g/kg) , once a day for 11 days. And in model group and normal control group, the mouse were gavage administrated 0.9% saline, once a day for 11 days. The uterine pathological sections of mice were conducted by HE and immunohistochemical method. The expression level of Ang 1, Ang 2 and Tie 2 protein in endometrium were detected by Mias 2000 image analysis system, and the endometrial thickness and numbers of uterine vessels were also measured. Result: Compared to that of normal control group, endometrial thickness and Ang 2 in the model group, Shou Tai Wan high, medium and low dose groups, and aspirin group had no any obvious change. The number of uterine vessel, and the expression level of Ang 1 and Tie 2 in the model group were significantly less than that of normal control group (P〈0.05). Compared to that of model group, the number of uterine vessel, expression level of Angl in high, medium or low dose Shou Tai Wan groups and aspirin group significantly increased (P〈0.05), and the Tie 2 expression level of rice endometrium in high or medium dose Shou Tai Wan groups and in aspirin group also significantly increased (P〈0.05). Conclusion: Shou Tai Wan improves endometrial receptivity of mice with COH through regulating expression of endometrial Ang and receptor, and increasing the number of uterine vessels.
