酒女贞子HPLC指纹图谱的建立及多成分含量的测定

摘　　要：目的:建立酒女贞子饮片的HPLC指纹图谱及建立指标成分含量测定的方法,为酒女贞子饮片的质量控制提供参考。方法:采用ODS C18(250 mm×4.6 mm,5μm)色谱柱,以乙腈-水为流动相,梯度洗脱,流速1.0 m L·min^(-1),检测波长230 nm,建立酒女贞子HPLC指纹图谱,并对红景天苷、齐墩果酸、木犀草素及特女贞苷指标成分的含量测定方法进行方法学考察。结果:红景天苷、齐墩果酸、木犀草素及特女贞苷线性范围分别为0.464~2.320、0.204~4.080、0.041~0.205和0.620~3.100μg,线性关系良好(r>0.999 8)。使用外标法测定红景天苷、齐墩果酸、木犀草素和特女贞苷含量分别在2.648~4.681、5.082~11.770、0.059~0.107和5.158~12.25 mg·g^(-1)范围内。取10个收集地酒女贞子饮片建立HPLC指纹图谱,以木犀草素峰为参比峰,样品相似度与对照图谱相似度均≥0.95,说明各地出售的酒女贞子饮片质量相对稳定。结论:该方法简便快速,分离效果好,重复性好,可为规范及评价酒女贞子质量及同属植物的研究提供参考。Objective： To establish an HPLC fingerprint method for the determination of ingredients of wine-fried Ligustri Lucidi Fructus, so as to provide a reference for the quality control. Methods： ODS C18 （ 250 mm × 4.6 mm, 5μm ） column was adopted, the mobile phase consisted of acetonitrile-water with gradient elution at the flow rate of 1.0 mL·min^-1, and the detection wavelength was 230 nm; the establishment of wine-fried Ligustri Lucidi Fructus HPLC fingerprint, and salidroside, determination method of oleanolic acid, luteolin and specnuezhenide index components were studied methodologically. Results： Good linear relationship were observed for Salidroside, oleanolic acid, luteolin and specnuezhenide in the ranges of 0.464 - 2.320, 0.204 - 4.080, 0.041 - 0.205 and 0.620 - 3.100 μg, respectively （ r 〉 0.999 8 ）. The content ranges of alidroside, oleanolic -1 acid, luteolin and specnuezhenide were 2.648-4.681,5.052-11.770, 0.059-0.107 and 5.158-12.25 mg·g^-1 by external standard method of determination. Different wine-fried Ligustri Lucidi Fructus were collected from 10 areas to establish HPLC fingerprint, and the luteolin was employed as a reference. The similarity of sample and standard drug is equal to 0.95 or higher, which indicated that the quality of wine-fried Ligustri Lucidi Fructus is relatively stable. Conclusion： The established method is simple, rapid, in good separation and reproducibility, and thus can provide a reference for the standard and evaluation of quality of wine-fried Ligustri Lucidi Fructus and congeneric plant.
