摘  要:采用流式细胞术研究离子液体1-丁基-3-甲基咪唑氯盐([C4mim][Cl])和1-丁基-3-甲基咪唑四氟硼酸盐([C4mim][BF4])对四尾栅藻的毒性影响,测定了细胞增殖、叶绿素荧光、细胞膜完整性和酯酶活性的变化。结果表明,2种离子液体均可抑制细胞增殖,且抑制作用随离子液体质量浓度和暴露时间增加而增强。计算得到96 h[C4mim][Cl]和[C4mim][BF4]的半数效应质量浓度EC50分别为49.61 mg/L和42.22 mg/L。测试期间发现,[C4mim][Cl](质量浓度20~200 mg/L)和[C4mim][BF4](质量浓度20~150 mg/L)对叶绿素荧光强度有一定的抑制作用,对细胞膜完整性无明显影响,随离子液体处理时间延长,对酯酶活性表现为先刺激后抑制作用;经200 mg/L[C4mim][BF4]处理藻细胞96 h后,藻细胞叶绿素的平均荧光强度仅为对照组的20%,酯酶活性为对照组的5%,且95%细胞膜发生破裂。2种离子液体对四尾栅藻均有毒性作用,这表明离子液体一旦进入水环境,将会对水生环境带来一定风险。This paper takes it as a goal to identify and determinate the toxic effects of two kinds of ionic liquids (ILs) [ C4mim] [ C1] and [C4mim][BF4] ) on the green microalgae Scenedesmus quadricauda, a key indicator of the aquatic environmental stress. For this purpose, we have made an investigation of the cell proliferation, the chloro- phyll fluorescence, the membrane integrity and the esterase activity of Scenedesmus quadricauda via the flow cytometer (FCM) . In our ex- perimental tests, S. quadricauda was let to be exposed to ILs in a range of mass concentrations from 0, 20 mg/L 50 mg/L, 100 mg/L, 150 mg/L to 200 mg/L. When the tests were over, all the samples were subject to an analysis in an interval of time period of O, 24 h, 48 h, 72 h and 96 h. Here, both the ILs should confine the cell proliferation of S. quadricauda, with the EC50 values of 49.61 mg/L and 42.22 mg/L for [C4mim] [C1] and [C4mim] [BF4] after 96 h exposure, respectively. In addition, all the inhibition rate for the cell proliferation should be made increasing with the increase of both the ILs concentration and incubation time, though the concentra- tion dependence on the chlorophyll fluorescence proves to be less ob- vious. After being exposed to [ C4mim] [C1] with mass concentration rangefrom 20 mg/L to 200 mg/L and [C4mim] [BF4] with mass concentration range from 20 mg/L to 150 mg/L for 96 h, the average chlorophyll fluorescence intensity is 34%- 39% and 28%- 41% lower than that of the control group, respectively; While after being exposed to 200 mg/L [C4mim][BF4] for 96 h, the average chloro- phyll fluorescence intensity is just 20 % as against the control group. After being exposed to 200 mg/L [C4mim] [BF4] for 96 h, the cell membranes of 95 % of the cells tend to get damaged with all the other samples being able to keep their membrane integrity.
