摘 要:本实验研究了6-BA、2,4-D、IAA、NAA、IBA、KT对蜡梅Chimonanthus praecox愈伤组织诱导的影响,以及探索愈伤组织诱导的最适培养基配方.激素筛选实验结果表明2,4-D能诱导蜡梅分化出愈伤组织.在2,4-D浓度梯度实验中,设计了0、0.1、0.25、0.5、1.0、1.5、2.0 mg/L 7个浓度,结果表明浓度为0.25~1.0 mg/L的2,4-D诱导愈伤组织的效果最好.在6-BA浓度梯度实验中,也设计了0、0.1、0.25、0.5、1.0、1.5、2.0 mg/L 7个浓度,均未诱导出愈伤组织,表明6-BA不能单独诱导出蜡梅愈伤组织.以葡萄糖、6-BA和2,4-D三因素组合正交实验共设置了25个组合,得到诱导蜡梅愈伤组织的最适培养基配方为:1%葡萄糖+0.25 mg/L2,4-D+1.5 mg/L6-BA.The purpose of this experiment was to study the effect on callus of moss Chimonanthus praecox made by 6-BA, 2,4-D, IAA, NAA, IBA, KT and the most suitable ratio of culture medium. The results of the experiment showed: 2,4-D can induce callus of Chimonanthus praecox. In the 2,4-D concentration gradient experiments, 0,0.1,0.25, 0.5,1.0,1.5,2.0 mg/L, a total of seven experimental concentration were set up. The results showed that the concentration of 0.25 - 1.0 mg / L of 2,4-D induced the largest callus with the largest area. In the 6-BA concentration gradient experiments, 0,0. 1,0.25,0.5, 1.0, 1.5,2.0 mg / L, a total of seven experimental concentration were set up, the results showed that 6-BA cannot induce callus of Chimonanthus praecox. Then, Glucose,6-BA and 2,4-D combined ratio for the three orthogonal factors. In these experiments, twenty-five experimental concentration were set up. The results showed that the combination ( 1% Glucose + 0.25 mg/L2,4-D + 1.5 mg / L6-BA) can induce the best callus.
