HPLC-ELSD法测定贝母二冬膏中西贝母碱的含量

摘　　要：目的：采用高效液相-蒸发光散射检测法测定贝母二冬膏中西贝母碱含量。方法：采用FORTISC18柱（4.6 mm×250 mm,5μm）,以乙腈：0.03%二乙胺溶液（25：75）为流动相,流速：1.0 m L·min-1,柱温：30℃,漂移管温度：67℃,气体流速2.0L·min-1。结果：西贝母碱在0.006-0.120 mg/m L浓度范围内有良好线性关系（r=0.9999）,平均加样回收率为97.9%,RSD%为1.42%。结论：本方法灵敏度高、准确、重复性好、专属性强,可作为控制贝母二冬膏质量的重要方法。Objective： To establish HPLC-ELSD method for the determination of Sipeimine in Beimu Erdong electuary. Methods： The HPLC system consisted of the FORTIS-C18 （4.6 mm×250 mm, 5 μm） column, the mobile phase consisted of acetonitrile （25） and 0.03% diethylamine （75）, flow rate was 1.0 mL·min-1 , the column temperature was 30 ℃, the drift tube temperature of ELSD was set at 67 ℃ with the flow rate of 2.0 L·min-1. Results： The linear response range was 0.006-0.120 μg/mL（r=0.9999）. The average recovery of Sipeimine was 97.9%, and RSD was 1.42%. Conclusions： The method was high sensitivity, accurate, repeatable and high secificity. It can be an important method to control the standard of Beimu Erdong electuary.
