摘 要:目的:研究甘油单油酸酯/泊洛沙姆F127(GMO/F127)溶致液晶作为控释药物的载体。方法:采用热处理法和乳化法,制备GMO/F127溶致液晶,研究了GMO/F127溶致液晶流体力学等物理特性。设计并完成溶致液晶担载不同极性药物体内和体外实验。采用分子模拟生物信息学方法研究了分子间相互作用。结果:制备了甘油单油酸酯/泊洛沙姆F127(GMO/F127)溶致液晶,无论对于水溶性药物还是脂溶性药物,GMO/F127溶致液晶都具有缓释作用,且对于2种药物的释放率都较高。脉冲电场控释对小鼠血糖影响的实验显示,胰岛素包裹的粒子具有良好的电场响应性。结论:GMO/F127溶致液晶粒径较小,同时具有较大的比表面积,能够作为一种优良的药物载体。GMO/F127溶致液晶担载药物的释放过程平稳而长效,并且能够在电场效应的刺激下将药物迅速释放出去。Objective: To research lyotropic liquid crystal as a kind of controlled release drug carrier. Methods : We investigated the preparation method of drug carrier and the physical characteristics of lyotropic liquid crystal. We completed experiments for different polarity drug loaded lyotropic liquid crystal particles in vivo and in vitro. We studied molecular interaction of lyotropic liquid crystal by molecular dynamic simulation. Results: GMO! F127 was synthesized by heat treatment method and emulsification method. GMO/F127 had a sustained release effect, and the release rate of the two drugs was high whether it was water soluble or lipid soluble. In mouse experiment, the effect of pulse electric field-controlled release on blood glucose showed that the particles had a good electric field response. Conclusion: Lyotropic liquid crystal is a wonderful drug carrier because of the smaller particle size and the greater specific surface. We found that the drug might be released steadily and long-acting. The release of drug molecules may be induced by pulsed electric field.
