摘 要:目的:建立HPLC-ELSD同时测定开心散提取物中主要抗抑郁类成分tenuifoliside A,1-O-(E)-benzoyl-[3-O-(E)-alphatolluyl]-β-D-fructofuranosy-(2→1)-[β-D-glucop-yranosyl-(1→2)]-α-D-glucopyranoside和人参皂苷Rb_1含量的方法。方法:采用HPLC-ELSD分析方法,Agilent HC C_(18)色谱柱(4.6 mm×250 mm,5μm),柱温25℃,流动相乙腈-0.65%乙酸铵水溶液进行梯度洗脱,流速1.0 m L·min^(- 1),漂移管温度115℃,载气流量3.2 L·min^(- 1),气体空气。结果:tenuifoliside A,1-O-(E)-benzoyl-[3-O-(E)-alphatolluyl]-β-D-fructofuranosy-(2→1)-[β-D-glucop-yranosyl-(1→2)]-α-D-glucopyranoside和人参皂苷Rb_1分别在1.113~10.980,1.581~15.726,0.712~7.128μg呈现良好的线性关系;精密度、稳定性和重复性均符合定量分析要求;三者的平均回收率分别为98.36%,99.58%和98.97%;百分含量分别为0.999%,1.670%和0.579%。结论:该分析方法简便、可靠,可用于开心散提取物中tenuifoliside A,1-O-(E)-benzoyl-[3-O-(E)-alphatolluyl]-β-D-fructofuranosy-(2→1)-[β-D-glucopyranosyl-(1→2)]-α-D-glucopyranoside和人参皂苷Rb_1的含量测定,且可以作为开心散质量控制方法之一。Objective: To found a quantitative HPLC-ELSD method for determining tenuifoliside A, 1- O- (E) -benzoyl- [3-0- (E) -alphatolluyl] β-D-fructofuranosy- (2 - 1) - [β-D-glucop-yranosyl- (1 2) ] -a-D-glucopyranoside and ginsenoside Rb1 from Kaixin San, which are used for anti-tumor. Method: The procedure of HPLC-ELSD was performed on the chromatographi column of Agilent HC C18 (4.6 mm x 250 mm, 5 μm)at 25 ℃ , and the mobile phase was acetonitrile-0.65% ammonium acetate solution in gradient elution. The flow velocity was 1.0 mL·min-1. The temperature of driti tube was 115℃ and the gas was air, flow rate was 3.2mL·min-1. Result: Tenuifoliside A showed a good linear relationship at a range from 1. 113 μg to 10. 980 μg, 1-0- (E) -benzoyl- [ 3-O- (E) -alphatolluyl ] -β-D-fructofuranosy- ( 2 - 1 ) [β-D-glucop-yranosyl- ( 1 2) ] -a-D-glucopyranoside, from 1. 581 p,g to 15. 726 μg, and ginsenoside Rba, from 0. 712μg to 7. 128μg; Precision, stability and repeatability are comply with the requirement of quantitative analysis. The average recovery was respectively 98.36% , 99.58% and 98.97%. The percentage composition was respectively 0. 999% , 1. 670% and 0. 579%. Conclusion: The method was simple and reliable, which can be used for the assaying of tenuifoliside A, 1-0- (E) -benzoyl-β-0- (E) -alphatolluyl ] -β-D-fructofuranosy- ( 2-→ 1 ) β-D-glucop- yranosyl- (1-→2)] -ol-D-glucopyranoside and ginsenoside RbI as well as quantity control of Kaixin San.
