摘 要:目的:探讨Foxp3和RORγt在肝泡型包虫病患者肝脏组织中的表达及意义。方法:将手术治疗的14位肝泡型包虫病患者的肝脏组织分为3组:病灶组织组(L组)14例,病灶旁组织组(P组)14例,正常肝组织组(N组)14例。应用实时荧光定量PCR(q RT-PCR)法检测各肝脏组织中Foxp3和RORγt m RNA的表达,分析其结果。结果:Foxp3 m RNA在L组中的表达明显高于P组(P=0.005)和N组(P=0.004),P组中的表达高于N组(P=0.246)。RORγt m RNA在L组中的表达高于P组和N组(L组vs P组,P=0.187);L组vs N组,P=0.044),在P组中的表达高于N组(P=0.027)。肝脏不同组织中,RORγt与Foxp3的表达量呈正相关。结论:Th17细胞转录因子Foxp3和Treg细胞转录因子RORγt在肝脏病灶组织中m RNA的表达升高,这些变化可能与泡型包虫病发病过程中寄生虫肉芽肿的形成有关。Objective: To investigate the expression and effect of Foxp3 and RORrt in liver tissue from Alveolar Echinococcosis patients. Methods: All samples were selected from 14 patients ith hepatic AE who were treated at the First Affiliated Hospital of Xinjiang Medical University.Diveded the tissues into 3 groups: 14 were lesion tissues(L), 14 were para-lesion tissues(P)and14 were normal liver tissues(N). The tissues expression levels of Foxp3 and RORrt gene were measured by real-time PCR(q RT-PCR). For statistical analysis, Student's t test and Pearson correlation were used. Results: The hepatic Foxp3 levels in the group L were significantly higher than that in group P(t=3.778, P=0.005)and group N(t=3.990, P=0.004). There was no significant difference between group P and group N(t=1.252, P=0.246), Meanwhile, RORγt gene expression levels in group P were also significantly higher than that in group N(t=2.705, P=0.027),but there was no significant difference between group L and group P(t=1.355, P=0.187). And there was also have significant difference between group L and group N(t=2.261, P=0.044). And the expression levels of RORγt and Foxp3 is positive correlate in different liver tissue. Conclusion: The expression of Th17 transcription factor RORrt and Treg transcription factor Fox P3 are increased in hepatic lesion, they may involve the process of parasitic granuloma formation.
