海马mGluRs的表达与大鼠空间学习记忆能力

摘  要:目的研究大鼠空间学习记忆能力与海马代谢型谷氨酸受体(m GluRs)表达相关性。方法将90只健康雄性大鼠随机分为空白对照组、小强度组和大强度组各30只,连续进行5 d Morris水迷宫训练,7 d后更换站台位置,再次进行5 d Morris水迷宫训练,每轮训练结束2 h后,取材检测海马CA1区m GluR1、m GluR5表达。结果小强度组及大强度组大鼠再次训练后平均逃避潜伏期均比首轮训练后显著缩短;小强度组和大强度组在首轮训练和再次训练后,CA1区m GluR1、m GluR5表达均明显增高;小强度组再次训练后海马CA1区m GluR1升高幅度明显比首轮训练低,大强度组再次训练后海马CA1区m GluR1表达差异无统计学意义;小强度组两轮训练海马CA1区m GluR5表达差异无统计学意义,大强度组再次训练后海马CA1区m GluR5升高幅度明显比首次训练高。结论大鼠空间学习记忆能力与m GLURs表达变化相关,其机制有待于进一步研究。Objective To research the relationship between the rat spatial learning and memory ability and expression of metabotropic glutamate receptors (mGluRs)in hippocampus. Methods 90 healthy male rats were randomly divided into blank control group, small intension training group, and large intension training group by Morris water maze with 30 rats, respectively. After 7 d of first 5 d training Morris water maze had been training again for 5 d through replacement platform position. After the end 5 d training period, the expression of mGluRl and mGluR5 has been detected in CA1 area of hippocampal by immunohistochemistry. Results The training average escape latency of rats in small intension training group and large intension training group after the second training was significantly shortened after the first training. The small intension training group and large intension training group after the training, mGluR1 and mGluR5 expression in CA1 area was significantly increased. The rising extent of mGluRl in small intension training group after the second training was obviously lower than the first training in hippocampal CA1 area, but non difference in large intension training group. The mGluR5 expression of small intension training group after two training was no significant difference in hippocampal CA1 area. However, the mGluRs expression after the second training was higher than that in the first training. Conclusion The same type training can improve the spatial learning and memory of rats through changing the mGluRs expression, but the mechanism need a further study.
