摘 要:目的观察绝经后骨质疏松症患者组蛋白去甲基化酶JMJD2家族的变化及其对组蛋白甲基化(H3K9me3、H3K36me3)水平的影响,揭示组蛋白去甲基化酶JMJD2家族与绝经后骨质疏松症的关系,为骨质疏松症的诊治提供新的思路。方法选择年龄50至70岁因髋关节骨性关节炎行关节置换的绝经后患者,通过检测腰椎骨密度,收集10例绝经后骨质疏松症患者(实验组)和10例非骨质疏松症患者(对照组),术中提取股骨头松质骨标本,免疫组织化学、蛋白质印迹法检测骨组织中组蛋白去甲基化酶(JMJD2A、JMJD2B)和组蛋白甲基化(H3K9me3、H3K36me3)表达。结果绝经后妇女JMJD2A、JMJD2B表达为弱阳性到阳性不等,骨质疏松症患者组表达水平低于非骨质疏松症患者组,组间差异具有统计学意义(P<0.05);骨质疏松症患者组H3K9me3、H3K36me3表达水平高于非骨质疏松症患者组,组间差异具有统计学意义(P<0.05)。结论绝经后骨质疏松症与组蛋白高甲基化状态相关,组蛋白去甲基化酶JMJD2A、JMJD2B能够降低骨质疏松的发生,组蛋白去甲基化酶JMJD2A、JMJD2B可能作为绝经后骨质疏松症的一种拮抗酶。Objective To study the expression of histone demethylase JMJD2 family in postmenopausal women with osteoporosis and its impact to histone methylation (H3K9me3, H3K36me3 ) levels, to establishment the links between histone demethylase JMJD2 family and postmenopausal osteoporosis, and to provide a new method for the prevention and treatment of osteoporosis. Methods Twenty 50-70-year-old postmenopausal women, who had THA due to osteoarthritis of the hip, were selected. After measurement of BMD of the lumbar spine, they were divided to study group ( 10 patients with osteoporosis) and control group ( 10 without osteoporosis). During THA, the cancellous bone specimen were collected from the femoral head. The expressions of JMJD2A, JMJD2B, H3K9me3, and H36me3 were detected using immunohistochemistry and Western blotting. Results JMJD2A and JMJD2B expression was from weak positive to positive in postmenopausal women. The expression level was lower in osteoporosis group than that in non-osteoporosis group, with a statistical difference (P 〈 0.05). The expression levels of H3K9me3 and H3K36me3 were higher in osteoporosis group than those in non-osteoporosis group, with a statistical difference (P 〈 0.05). Conclusion Postmenopausal osteoporosis is associated with the histone methylation level. The histone demethylase JMJD2A and JMJD2B reduce the occurrence of osteoporosis. Histone demethylase JMJD2A and JMJD2B may serve as antagonistic enzymes of postmenopausal osteoporosis.
