摘 要:采用溶胶-凝胶法制备了细胞色素c/二氧化硅纳米粒,通过透射电镜、紫外-可见吸收光谱、荧光光谱、同步荧光荧光光谱等多种检测手段分别对细胞色素c(Cytc)与包裹后的细胞色素c(Cytc SN)结构和性质进行了表征。研究表明,Cytc SN外形为球形粒子,在溶液中具有很好的单分散性及稳定性。紫外-可见吸收光谱显示Cytc SN仍然保持了Cytc的血红素辅基特征吸收Soret吸收带,但使Cytc SN的构象发生变化。分别对Cytc与Cytc SN的储藏稳定性、热稳定性与对有机溶剂耐受稳定性进行比较。结果表明:放置2 d后,Cytc SN的溶液依旧澄清透明;而Cytc由于蛋白质聚合作用,溶液出现浑浊。从10℃加热到40℃的过程中,Cytc与Cytc SN的吸光度变化率不大,当温度高于40℃后,Cytc的结构发生明显变化,而Cytc SN则显示出更优的热稳定性。此外,Cytc SN对有机溶剂的耐受性与Cytc相比有所提高。In this paper,cytochrome c / silica nanoparticles have been prepared by sol-gel method. The structural properties and character of Cytc and Cytc SN were characterized and analyzed by transmission electron microscopy( TEM),ultraviolet-visible spectroscopy( UV-Vis),infrared spectroscopy( IR) and synchronous-scan fluorescence spectroscopy( SFS). The results indicated that Cytc SN can stable disperse in aqueous solution for a long time. After encapsulated inside silica nanoparticles,the formation of Cytc was partly changed but the Soret band was maintained.Cytc SN can well disperse in aqueous solution for more than two days and showed little structure changing even heated higher than 40℃. Besides,the ability for tolerant organic solvent of Cytc SN was higher than that of Cytc.
