摘 要:目的探讨瘦素基因rs2167270、rs4731426和瘦素受体基因rs12405556单核苷酸多态性与中国南方汉族人群冠心病(Coronary Artery Disease,CAD)的遗传易感性的关系。方法应用SNPscan TM多重SNP分型试剂盒对中国南方汉族人的1 044例冠心病患者和1 349例健康对照样本进行SNP分型;等位基因关联分析采用卡方检验;基因型关联分析采用单因素和多因素非条件Logistic回归分析方法。结果等位基因和基因型关联分析未发现rs2167270、rs4731426和rs12405556与冠心病有关联(P=0.242~0.978);进一步按性别、是否患高血压、是否患2型糖尿病这3个混淆因素分层后,仍未发现3个SNP在各亚组与冠心病有关联(P=0.0938~0.997)。此外,也未发现这3个SNP与冠心病合并高血压/2型糖尿病存在显著关联(P=0.258~0.994)。结论本研究未发现瘦素基因rs2167270、rs4731426及瘦素受体基因rs12405556单核苷酸多态性与中国南方汉族人群的冠心病有关联。Objective The aim of this study was to assess the association of single nucleotide polymorphisms (SNP) in leptin (rs2167270, rs4731426) and leptin receptor (rs12405556) with coronary artery disease (CAD) in a southern Chinese Han population. Methods Genotyping for 1044 CAD patients and 1349 healthy individuals of southern Chinese Hart were conducted by SNPscanTM multiple SNP genotyping assays. Allelic association was assessed using Chi-square test. And genotypic association analysis was performed by unconditional logistic regression models. Results Both allelic and genotypic association analysis did not reveal any significant association between the three SNPs (rs2167270, rs4731426 and rs12405556) and CAD (P=0.242~0.978). Further stratification analysis by three confounding factors (gender, hypertension or type 2 diabetes mellitus) also did not show any significant (allelic and genotypic) association of the three SNPs with CAD in these subgroups (P=0.0938~0.997). In addition, there was no significant association between the three SNPs and CAD syndromes (complicated either with hypertension or type 2 diabetes mellitus (P=-0.258~0.994). Conclusion The study did not reveal any significant association between three SNPs (rs2167270, rs4731426 in LEP or rs12405556 in LEPR) and CAD in the southern Chinese Han population.
