口服灭活幽门螺杆菌对新生BALB/c小鼠胃黏膜的免疫保护作用研究

摘  要:目的:探索BALB/c小鼠新生期口服灭活幽门螺杆菌(H.pylori)后对胃黏膜是否具有免疫保护作用。方法:将32只BALB/c小鼠分为四组进行实验。第Ⅰ组为PBS对照组,第II组为灭活H.pylori组,第Ⅲ组为灭活H.pylori加大肠杆菌不耐热肠毒素B亚单位(LTB)组,第IV组为正常对照组。用相应抗原成分对Ⅰ、Ⅱ、Ⅲ组出生24 h以内的新生BALB/c小鼠进行灌胃免疫,末次免疫两周后,将小鼠用108CFU的幽门螺杆菌SS1株进行活菌灌胃攻击,连续攻击两天,每天两次。在攻击后的第28天,将各组小鼠处死,取胃组织进行活菌计数及组织病理学炎性评分。结果:Ⅰ、Ⅱ、Ⅲ、Ⅳ组慢性炎症评分分别为2.25±0.46、2.12±0.35、1.5±0.53、0.25±0.46,炎症活动度评分为2.12±0.35、2.12±0.35、1.37±0.52、0.25±0.46;除Ⅰ、Ⅱ组间无统计学意义外,其余组间差异均有统计学意义(P<0.05)。Ⅰ、Ⅱ、Ⅲ组细菌定植量分别为1.33±0.21、1.18±0.27、1.05±0.37×105CFU/1个胃,Ⅳ组细菌定植量<4×102CFU/1个胃,低于检测值,Ⅲ组的细菌计数低于Ⅰ、Ⅱ组,差异有统计学意义(P<0.05)。结论:BLAB/c小鼠新生期给予灭活幽门螺杆菌加LTB灌胃免疫与活菌攻击后炎症的减轻和细菌定植量的减少可能相关,具有一定的免疫保护作用。Objective: To explore the Immunoprotection of gastric mucosa in newborn mice after oral immunization with inactived H. pylori. Methods: Thirty-two BALB/c mice were randomly divided into four groups, including PBS control group (group I ), the group of inactivated H. pylori ( group Ⅱ ), the group of inactivated H. pylori and Escherichia cole heat-labile enterotoxin B subunit ( LTB ) ( group Ⅲ ), and normal group ( group Ⅳ). Newborn BLAB/c mice within 24 hours were immunized through oral gavage with corresponding antigen,Two weeks after final immunization, the mice were challenged with 1 × 10SCFU live H. pylori strain SS1, twice once day ,for 2 days. The twenty-eighth day post-challenge,all of mice were sacrificed and stomachs were removed for grades of inflammation and The counting of H. pylori colonizing. Results: The mean scores of stomach tissue inflammation of group I ,Ⅱ , Ⅲ and IV were 2. 25 ±0.46,2. 12±0. 35,1.5 ±0. 53,0. 25 ±0.46, respectively, and the score of inflammatory activity of the above four groups were 2. 12±0. 35,2. 12±0. 35,1.37 ±0. 52,0.25 ±0.46. Among these four groups, the results were statistically significant between any two groups except the result between group I and group Ⅱ Finally, H. pylori colonization quantity in the stomachs of group HI was lower than group I and groupⅡ (P〈 0. 05),as expected,the H. pylori colonization was undeteetable in group IV. Conclusion: The oral garage with inaetived H. pylori plus LTB to newborn mice may be associate with the lightening of inflammation and the decrease of H. pylori colonizing number.
