摘  要:为提高卵母细胞体外成熟及孤雌胚胎发育的能力,探讨了双丁酰基腺苷酸环化酶(db-c AMP)和卵丘细胞单层共培养对猪卵母细胞体外成熟及孤雌胚胎体外发育潜力。结果表明,成熟培养液中分别添加0、0.5、1.0、1.5和2.0 mmol/L的db-c AMP时,卵母细胞的体外成熟率依次降低,但浓度为1 mmol/L时的孤雌胚囊胚率(27.03%)显著高于其他各组(P<0.05);卵丘细胞单层共培养卵母细胞时,其成熟率、卵裂率及囊胚率(86.00%、89.04%、28.36%)也得到了显著提高;0.4 mmol/L的db-c AMP和卵丘细胞单层共培养时,其成熟率和卵裂率(85.78%和88.95%)与对照组差异不显著,但囊胚率(35.14%)显著提高。0.4 mmol/L的db-c AMP和卵丘细胞单层共培养能够显著提高猪卵母细胞的体外成熟及孤雌胚胎的发育潜力。To improve the developmental potential of pig oocytes in vitro maturation and parthenogenetic embryos, the effects of db-cAMP concentration and cumulus cells monolayer on in vitro maturation and parthenogenetic potential of pig oocytes was investigated. The tests indicated that as the concentration of db-cAMP increased (0、0.5、1.0、1.5 and 2.0 mmol/L), the maturation rate of oocytes reduced successively. But the blastocyst rate of 1 mmol/L group (27.03%) was significant higher than others(P﹤0.05); When oocytes were co-cultured with pig cumulus cell monolayer, the maturation rate、cleavage rate and blastocyst rate (86.00%、 89.04%、 28.36%) were significantly improved. 0.4 mmol/L db-cAMP co-cultured with the monolayer cumulus cell had no significant difference in maturation rate and cleavage rate (85.78% and 88.95%), but the blastocyst rate (35.14%) were significantly improved. 0.4 mmol/L db-cAMP co-cultured with monolayer cumulus cell can improve the development of parthenogenetic embryos.
