摘  要:目的探讨梓醇、左旋紫草素、丹皮酚通过影响JNK信号通路发挥调控HBD-2的作用。方法用IL-1β和TNF-α各100ng/m L的混合物刺激Ha Ca T细胞18h建立HBD-2高表达及JNK信号通路活化的细胞学体外模型,配制含合适浓度的梓醇、左旋紫草素、丹皮酚培养基干预细胞模型,并设阴性及阳性对照。用实时荧光定量PCR法检测各组中HBD-2 m RNA的表达;用ELISA法检测培养上清中HBD-2蛋白的表达;用Western blot法检测各组中JNK蛋白的表达。结果左旋紫草素、丹皮酚和JNK抑制剂(阳性对照药物)可以明显下调细胞学体外模型中HBD-2基因及蛋白的表达(P<0.05),左旋紫草素与JNK抑制剂相当(P>0.05),丹皮酚弱于JNK抑制剂(P<0.05)。梓醇则明显上调细胞学体外模型中HBD-2基因及蛋白的表达(P<0.05)。左旋紫草素、丹皮酚和JNK抑制剂可以明显下调P-JNK2蛋白的相对含量,梓醇对P-JNK2蛋白无抑制作用,反而上调其表达。结论紫草、丹皮可能通过抑制JNK2MAPK信号通路的活化,下调HBD-2的表达,从而发挥其治疗银屑病的作用。地黄治疗银屑病的机制尚需从其他方面继续探讨。Objective To observe the influence of catalpol,L-shikonin and paeonol in regulating the HBD-2 expression in keratinocyte by JNK signaling pathway. Methods Ha Ca T cells were stimulated by a mixture of 100 ng / m L of IL-1β and TNF-α for 18 h to establish HBD-2 high-expression and JNK signaling pathway activation in vitro model. Catalpol,L-shikonin and paeonol solution with appropriate concentration were prepared and added into the cell culture media directly. JNK inhibitor was selected as the positive control drugs. Effects of the three monomer of Chinese traditional herb on HBD-2 m RNA expression in the cell model were detected by real-time PCR. HBD-2 expression of the cell culture supernatant was detected by ELISA in each group. Effects of the three monomers of Chinese traditional herbs on JNK signaling pathway was detected by Western blot. Results L-shikonin,paeonol and JNK inhibitor could markedly decrease the expression of HBD-2 in vitro cytology model of psoriasis( P 〈0. 05). No significant difference was decteted between Lshikonin and JNK inhibitor group( P〉0. 05). The effect of paeonol was weaker than JNK inhibitors( P 〈0. 05). Catalpol could markedly increase the high expression of HBD-2 in vitro cytology model of psoriasis( P 〈0. 05). L-shikonin and paeonol could significantly decrease the relative content of P-JNK2 protein. Catalpol could significantly increase the relative content of P-JNK2 protein. Conclusion Lithospermum and cortex moutan may reduce the expression of HBD-2 through inhibit JNK2 MAPK signaling pathway activation in keratinocyte,and thus play the role in the treatment of psoriasis. The mechanism of Rehmannia glutinosa Libosch for the treatment of psoriasis still need exploration from other aspects.
