摘  要:目的观察平哮饮对哮喘模型小鼠氧自由基代谢的影响,探讨其作用机制。方法将60只雌性BALB/c小鼠随机分为6组,每组10只,分别为空白对照组、哮喘模型组、阳性(地塞米松)对照组和中药治疗组(平哮饮高、中、低剂量组)。除空白对照组外,其余各组用改良的卵白蛋白多点注射致敏与雾化吸入激发法建立小鼠哮喘模型。空白对照组注射和雾化吸入溶液均为生理盐水;阳性对照组从激发当天开始给予地塞米松;平哮饮组分别给予4、8、16 g/kg提取物,各组连续给药14 d。各组小鼠末次激发24 h后,处死各组小鼠留取标本。镜下观察各组哮喘小鼠肺组织病理形态变化,采用双抗体夹心酶联免疫吸附试验(ELISA)法测定小鼠肺组织匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)含量。结果平哮饮高、中剂量组可明显升高哮喘模型小鼠肺组织中SOD水平(P<0.05),降低MDA和NO含量(P<0.05)。平哮饮可明显减轻哮喘模型小鼠肺组织细胞的炎性浸润。结论平哮饮可能通过调整炎症状态下肺组织氧自由基水平,减轻氧化损伤程度,从而改善哮喘的气道炎症。Objective To observe the effect of Pingxiao Decoction on the metabolism of oxygen free radicals in mice model of bronchial asthma. Methods Sixty female BALB / c mice were randomly divided into 6 groups,blank control group,asthma model group,positive( dexamethasone) control group and Pingxiao Decoction treatment group( high,middle,low dose),each group with 10 mice. All the groups which Modified ovalbumin multipoint injection with aerosol inhalation method was used to establish asthma mice model besides the blank control group. Blank control group mice were injected and inhaled with saline. From the first sensitized day,positive control group mice were given dexamethasone and drug treatment groups mice were given 4,8,16 g / kg Pingxiao Decoction extract for 14 consecutive days. 24 hour after the end of sensitization,the mice were executed and lung tissue specimens were prepared. Lung tissue morphological of the mice changes in different groups were observed under microscope. Superoxide dismutase( SOD),malondialdehyde( MDA)and nitric oxide( NO) in lung tissue homogenate were assessed by enzyme linked immunosorbent assay( ELISA). Results The SOD level in high,medium dose groups of Pingxiao Decoction were significantly increased in lung tissue of asthmatic mice( P 〈0. 05),meanwhile MDA and NO levels were decreased( P 〈0. 05). Pingxiao Decoction could significantly reduce inflammatory cell infiltration in the lung tissue of asthmatic mice model. Conclusion Pingxiao Decoction could ameliorate the airway inflammation in asthma mice model by adjusting the oxidation and antioxidant system in vivo possibly.
