人脐血间充质干细胞两种移植方法在骨折模型中比较研究

摘　　要：目的：观察通过耳缘静脉和骨折局部注射，将人脐血间充质干细胞移植到新西兰兔骨折模型中的移植效果差异，为人脐血间充质干细胞临床应用提供依据。方法建立新西兰兔胫骨横断骨折模型，随机分为经骨折局部移植组（A 组）和耳缘静脉移植组（B 组）。在骨折模型建立后第3天，A 组注射浓度为1＆#215;107个/ m1、经 BrdU 标记的人脐血间充质干细胞悬液1 m1；B 组注射等量经 BrdU 标记的人脐血间充质干细胞悬液。细胞移植后24、48、72 h 观察兔大体情况、取骨折断端组织行免疫荧光显微镜观察、HE 染色光镜观察。结果细胞移植后 A、B 两组大体情况观察无明显差异；A、B 两组免疫荧光显微镜均可见 BrdU 标记的阳性细胞，但 A 组阳性细胞计数明显多于 B 组，两组比较，差异有统计学意义（P 〈0.05）；两组 HE 染色光镜观察，A 组炎细胞计数明显多于 B 组，两组比较，差异有统计学意义（P 〈0.05）。结论人脐血间充质干细胞经耳缘静脉和骨折局部移植均是安全可行的，但经骨折局部注射移植更适合应用于骨折模型而有利于骨创伤修复。Objective To investigate the transp1antation of human umbi1ica1 cord mesenchyma1 stem ce11s into the fracture mode1 by ear vein injection and 1oca1 injection and then observe differences in transp1antation. To provide evidence to the c1inica1 app1ication of human umbi1ica1 cord b1ood mesenchyma1 stem ce11s. Methods The fracture mode1s were made and were random1y divided into ce11 1oca1 injection group（A group）and ear vein injection group（B group）. HUMSCs of BrdU-1abe1ed at a concentration of 1 ＆#215;107 ce11s/ m1 were respective1y injected into the two groups at the 3rd day when the mode1s were made. The rabbits were observed at 24 hours,48 hours and 72 hours after ce11 transp1antation for their genera1 reactions and took the fracture fragments tissues by immunof1uorescence microscopy and HE staining 1ight microscopy. Results There was no significant difference observed in the genera1 situation between the two groups. BrdU-positive ce11s were seen by immunof1uorescence microscopy in both two groups. However,the positive ce11s in group A were higher than group B and the difference was statistica11y significant（P 〈 0. 05）. HE staining 1ight microscopy showed that inf1ammatory ce11s in A group were higher than B group,the difference was statistica11y significant（P 〈 0. 05）. Conclusion The two ways of transp1antion of Human umbi1ica1 cord b1ood mesenchyma1 stem ce11s are safe and feasib1e. However,the 1oca1 injection in frac-ture is more suitab1e for transp1antation and is conducive to bone wound hea1ing.
