醉茄素-A抗肺鳞癌细胞上皮间质转化的相关研究

摘  要:目的探讨醉茄素-A(WFA)对肺鳞癌细胞上皮间质转化(上皮标志物E—cadherin和间质标志物Vimentin的表达)的影响,以及Notch受体拮抗剂对肺鳞癌细胞上皮间质转化(EMT)的作用及抗肺鳞癌的作用机制。方法体外培养肺鳞癌细胞(SK—MES-1)。用终浓度分别为0(对照组),10,20,40μg/mL的醉茄素-A组处理SK—MES-1细胞24h后,于相差显微镜下观察细胞的一般形态结构、脱落情况等;M1Tr法检测细胞的生长活性;酶联免疫法(ELISA)和RT—PCR法检测上皮标志物E—cadherin和间质标志物Vimentin的表达。结果不同浓度的醉茄素-A均能抑制SK—MES-1细胞增殖,其抑制率与作用浓度相关;ELISA结果提示,不同浓度WFA作用SK—MES-1细胞24h后,与对照组比较,浓度为20,40μg/mLWFA组,E—cadherin较对照组表达增加,与浓度呈正相关:Vimentin较对照组表达减少,差异有统计学意义(P〈0.05);其余浓度WFA组无明显变化;RT—PCR结果显示,在质量浓度为20,40仙g/mL醉茄素-A试验组E—cadherinmRNA表达增加,VimentinmRNA表达减少,差异有统计学意义(P〈0.05)。结论醉茄素-A能抑制肺鳞癌细胞的生长并抑制其上皮一间质转化(EMT)过程,醉茄素-A可能通过抑制肺鳞癌细胞发生上皮间质转化从而抑制肿瘤生长。Objective To investigate about the relationship between withaferin-A treat lung squamous carcinoma cell and Epithelial Senchymal transition(EMT); To study the effect of Notch receptor antagonist on epithelial mesenchymal transition (EMT) and the possible mechanism of anti lung squamous cell carcinoma.Methods Culture lung adenocarcinoma(SK-MES-1 )in vitro,and respec- tively use WFA final coneentratino 0 (control group),10,20,40 Ixg/mL group to cells after 24 h,then the cells of structure and loss situation were observed by the phase contrast microscope morphological structure; Determined by MTT method to detect the cell growth activity; the expression of Vimentin and E-cadherin were detected by enzyme linked immunosorbent assay(ELISA) and RT-PCR assay.Results The concentration of 10, 20, 40 μg/mL of withaferin-A can inhibit proliferation of SK-MES-1 cells, the inhibition rate relat to the action concentrations; ELISA results indicated that the expression of E-cadherin was signifi- cantly lower than that of control group, and the expression of Vimentin was significantly increased compared with the control group at different concentrations 20 I^g/mL and 40 g/mL WFA, respectively.The results RT-PCR was that the concentration of 20 and 40 μg/mL WFA in group E-cadherin mRNA expression increased, Vimentin mRNA expression reduced,the difference was statistically significant(P 〈 0.05).Conclusion WFA can inhibit the growth of lung squamous cancer cells and inhibit the ep- ithelial-interstitial conversion process,its occurrence relate to the raise of expression about E cadherin protein and gene,and the downgrade of expression Vimentin protein and gene.So we speculate that withaferin-A might inhibit lung squamous epithelial mes- enchymal changes of the cancer cells(EMT),thus inhibiting tumor growth, providing new thought and way for clinical treatment of lung cancer.
