摘 要:目的比较亚硫酸氢钠穿心莲内酯和硫酸卡那霉素对HK-2细胞毒性作用的差异。方法通过倒置显微镜观察亚硫酸氢钠穿心莲内酯和硫酸卡那霉素作用后HK-2细胞形态的改变,应用噻唑蓝(M1Tr)比色法和流式细胞术法检测药物对HK-2细胞的形态、细胞周期、凋亡率、线粒体膜电位(MMP)和活性氧(ROS)的影响。结果亚硫酸氢钠穿心莲内酯对HK-2细胞作用后24h和48h的‰值为(19.12±3.55)和(n.56±3.881)玑;硫酸卡那霉素对HK-2细胞的作用后24h和48h的IC50值为(119.35±6.22)和(120.25±22.09)g/L,随着硫酸卡那霉素作用的时间增加,IC50值变化不显著。西药对HK-2细胞的形态、细胞周期、凋亡率、MMP和ROS的影响不同.联合用药对细胞的损伤作用更大。结论亚硫酸氢钠穿心莲酯和卡那霉素可能以不同的途径对HK-2细胞产生毒性,联合作用可加重对HK-2细胞毒性作用。Objective To compare the differences of drug combination concerning andrographolidi natrii bisulfis and kanamycin sulfate on HK-2 cytotoxicity.Methods The morphological changes of andrographolidi natrii bisulfis and kanamycin sulfate on HK- 2 cells were examined with contrast microscopy and growth inhibitory rate of HK-2 ceils were detected by MTT color/metric as- say and the cell-cycle arrest, apoptosis,mitochondrial membrane potential(MMP)and reactive oxygen species(ROS) were analyzed by flow cytometry.Results The IC50 of andrographolidi natrii bisulfis and kanamycin sulfate against HK-2 cells for 24 and 48 hours were (19.12±3.55), (11.56±3.881 ), (119.35±6.22)and (120.25±22.09)g/L respectively.The IC5o of kanamycin sulfate didn't change significantly with increase of time. And there were lots of differences in HK-2 cells morphology,cell cycle,MMP,and ROS respectively treated with andrographolidi natrii bisulfis and kanamycin sulfate and their combination induced greater injury on HK-2 cells than the two drugs alone.Conclusion Andrographolidi natrii bisulfis and kanamycin sulfate might act on HK-2 ceils in different pathways to produce toxic effects and their combined effect can exacerbate the more venenous on HK-2 cells.
