补肾壮骨胶囊对兔膝关节关节软骨组织中基质金属蛋白酶及抑制因子的影响

摘　　要：目的研究补肾壮骨胶囊对兔膝关节关节软骨组织MMP-1，TIMP-1，ADAMTS-4的影响。方法选取体重为2．5妇～3．0奴的普通级新西兰大白兔44只，使用随机数表法将所有兔子随机均分为4组，第1组为造模组，第2组为观察组，第3组为对照组，第4组为正常组。对给药后各组兔膝关节关节软骨组织MMP-1．TIMP-1，ADAMTS-4进行比较。结果观察组和对照组的软骨组织中MMP-1阳性细胞分数无显著性差异（P〉0．05）；观察组和对照组的软骨组织中MMP-1阳性细胞分数显著低于造模组（P〈0．05）；观察组和对照组的软骨组织中TIMP-1阳性细胞分数无显著性差异（P〉0．05）；造模组的软骨组织中TIMP-1阳性细胞分数略高于正常组，两者无统计学差异（P〉0．05）；观察组和对照组的软骨组织中TIMP-1阳性细胞分数显著高于造模组和正常组（P〈0．05）；观察组和对照组的软骨组织中ADAMTS-4阳性细胞分数无显著性差异（P〉0．05）；正常组关节软骨组织中ADAMTS-4阳性细胞分数显著低于造模组（P〈0．05）；观察组关节软骨组织中ADAMTS-4阳性细胞分数显著高于正常组（P〈0．05），但低于造模组（P〈0．05）；对照组关节软骨组织中ADAMTS-4阳性细胞分数显著高于正常组（P〈0．05），但是低于造模组（P〈0．05）。结论补肾壮骨胶囊能够有效激活TIMP-1。抑制MMP-1及ADAMTS-4的表达，进而促进兔膝关节关节软骨组织的修复，对膝骨性关节炎的治疗研究具有重要的意义，值得进行进一步研究。Objective To study the effect of Bushen Zhuanggu Capsule on rabbit knee articular cartilage tissue MMP-1, TIMP-1, ADAMTS-4.Methods Weight of 2.5 to 3.0 kg were New Zealand rabbits were 44 as the research object,using a random nmnber table method all rabbits were randomly divided into 4 groups,group 1 modules to build, the two groups as the observation group,group 3（control group）,group 4 for the normal group. To give the medicine after each rabbit knee articular cartilage tissue MMP-1,TIMP-1,ADAMTS-4 were compared.Results In the observation group and the control group of cartilage tissue MMP-1 There were no significant difference in positive cell fraction（P〉 0.05）.The positive cell fraction of MMP-1 positive cells in obser- vation group and control group was significantly lower than that in model group （P〈 0.05）.The positive cells of TIMP-1 positive cells were significantly higher than that in normal group （P 〉 0.05）.The positive cell fraction of TIMP-1 in the cartilage tissues was significantly higher than that in normal group（P〈 0.05）.And control group of cartilage tissue ADAMTS-4 positive cell fraction of no significant difference （P 〉 0.05）;normal articular cartilage tissue fraction of ADAMTS-4 positive cells was significantly lower than that of modeling group（P 〈 0.05）;observation group,articular cartilage tissue fraction of ADAMTS-4 positive cells was significantly higher than that in the normal group（P 〈 0.05 ）,but less than modeling group（P 〈 0.05）;control group,articular cartilage tissue fraction of ADAMTS-4 positive cells was significantly higher than that in the normal group （P 〈 0.05）,but lower than the modeling group （P 〈 0.05）.Conclusion Kidney Zhuanggu capsule can The effective activation of TIMP-1 and the inhibition of MMP-1 and ADAMTS-4 expression,and then promote the repair of articular cartilage of rabbit knee joints,the research on the treatment of knee osteoarthritis has important significance,it is worth further research.
