摘 要:目的:观察通络救脑注射液及其有效组分对拟缺血损伤脑微血管内皮细胞谷氨酸受体3(glutamate receptor 3,GluR3)表达及其功能的影响。方法:采用大鼠原代培养脑微血管内皮细胞,氧糖剥夺法制备拟缺血模型,分别用通络救脑注射液及其2组分栀子苷、三七总皂苷对细胞进行干预,采用PCR和免疫蛋白印迹法检测各组细胞中GluR3 mRNA及蛋白表达水平,并用免疫蛋白印迹法检测GluR3调节的下游分子基质金属蛋白酶诱导因子CD147、基质金属蛋白酶9(matrix metalloproteinase-9,MMP-9)的蛋白表达情况。结果:通络救脑注射液及其组分能够从基因到蛋白水平显著下调GluR3的表达,对CD147、MMP-9也有显著下调作用,且通络救脑注射液疗效优于2组分。结论:通络救脑注射液及其组分可调节拟缺血损伤脑微血管内皮细胞GluR3的表达及其功能,这可能是其在脑缺血性损伤中保护血脑屏障、减轻炎症反应的机制之一。通络救脑注射液效果优于2组分,显示了复方配伍发挥整合调节的疗效优势。Objective: To study the effects of Tong Luo Jiu Nao injection (TLJN) and its active components on the expression and function of Glutamate receptor 3 (GluR3) in mimic ischemic brain microvascular endothelial cells (BMECs). Methods: Primary rat BMECs were used and mimic cerebral ischemia in vitro was established by oxygen-glucose-deprived (OGD) method. OGD-induced BMECs were treated with Geniposide,Panax notoginseng saponins (PNS) and TLJN respectively. The expression of GluR3 in mRNA and protein level was detected using RT-PCR and Western blot methods. The expression of CD147 and matrix metalloproteinase-9 (MMP-9) in various groups were detected by Western blot. Results: The expression of GluR3 in OGD-induced BMECs was down-regulated by TLJN and its components geniposide, PNS. The expression of CD147 and MMP-9 were also decreased in administration groups. Effect of TLJN was superior to geniposide or PNS. Conclusion: TLJN and its components could regulate the expression and function of GluR3 in mimic ischemic BMECs, which may be the internal mechanism of TLJN ' s effect on Blood Brain Barrier damage and inflammation in cerebral ischemia. Meanwhile, TLJN had more optimized effect than its active components, which showed the therapeutic advantages of Chinese medic/he compatibility.
