摘  要:目的:初步分析大黄总蒽醌胶囊含药血清中药源性成分。方法:大鼠灌胃给予大黄总蒽醌胶囊内容物,采集含药血清,高效液相色谱法测定其指纹图谱,与相同条件下测定的胶囊内容物、对照品及空白血清图谱比较,分析大鼠给药后体内产生的药源性成分并进行鉴定。结果:含药血清中产生药源性成分20个,其中6个为体外原形成分,14个为代谢产物,6个原形成分分别被鉴定为大黄素-8-O-葡萄糖苷、芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚,1个代谢产物被鉴定为芦荟大黄素-8-O-葡萄糖苷,其余13个代谢产物均具有大黄蒽醌类成分的紫外吸收特征。结论 :本研究建立的方法准确可靠,可用于大黄总蒽醌胶囊血清药化学研究。OBJECTIVE To preliminarily analyze drug-induced constituents in rats after oral administration of total rhubarb anthraquinone capsules (TRAC). METHODS Rat serum containing drug was prepared after intragastrie administration of TRAC. HPLC fingerprint of serum containing drug was detected by HPLC. By comparing fingerprints of serum containing drug and contents of TRAC, reference substances and blank serum, drug-induced constituents in blood were analyzed and identified. ~TS Twenty drug-induced constituents were detected in rat serum after oral administration of TRAC, six of them were original compounds contained in TRAC and fourteen were metabolites. Six original compounds were identified as emodin-8-glu- coside, aloe-emodin, rhein, emodin, chrysophanol, and physcion, respectively. One metabolite was identified as aloe-emodin-8-O- glueopyranoside. Other thirteen metabolites possessed the same ultraviolet absorption characteristics as those of anthraquino- nes. CONCLUSION The established method is accurate and reliable, can be used for studies on serum pharmaeochemistry of TRAC.
