摘 要:目的:建立脾虚小鼠原位移植肝癌模型,探讨脾虚对小鼠肝癌成瘤率和生存率的影响。方法:60只6周龄C57BL/6雄性小鼠随机分为正常组、脾虚组、脾虚肝癌组和肝癌组,每组15只。采用利血平建立小鼠脾虚模型,利血平皮下注射14天后,在脾虚模型基础上,hePal-6小鼠肝癌细胞经裸鼠皮下种植后移植入C57BL/6小鼠肝脏,制作肝癌模型并观察35天。实验过程中观察动物脾虚程度、成瘤情况及生存时间等。结果:脾虚组和脾虚肝癌组小鼠脾虚积分较正常组明显升高(P<0.01),平均分值分别为17.57±4.86和18.13±4.53,符合典型脾虚模型的分值范围。小鼠肝癌总的成瘤率可以达到86.67%。与肝癌组小鼠比较,脾虚肝癌模型组小鼠的成瘤率为100.0%,高于肝癌组的73.33%(P<0.05)。脾虚肝癌组小鼠的生存时间明显低于肝癌组小鼠(P<0.01)。结论:脾虚小鼠原位移植肝癌模型为从中医脾虚因素研究肝癌提供了简单可行的动物模型,脾虚是肝癌形成和预后不良的重要因素。Objective: To establish an orthotopic liver transplantation model in mice with spleen deficiency, and to explore the effect of spleen deficiency on the rate and survival rate of liver cancer in mice. Methods: sixty Mice were randomly divided into the normal group, the spleendeficiency group, liver cancer group and liver cancer after spleen-deficiency model group.The number of mice in Each group was 15 After the spleen-deficiency group and liver cancer after spleen-deficiency model group were given reserpine subcutaneous injection for 14 days, orthotopic transplantation liver cancer models were made. Li-ver cancer models were observed for 35 days. Degree of spleen-deficiency, survival time and tumor formation were recorded. Results: Spleen-deficiency mouse models were successfully established.The tumor forma- tion rate was 73.33% in the liver cancer model group and 100% in the liver cancer after spleen-deficiency model group (P〈0.05). The average survival time of the liver cancer group was higher than that of the liver cancer after spleen-deficiency model group (P〈0.01). Conclusion: The spleen-deficiency mouse model of orthotopic trans-plantation liver cancer provides a simple and feasible model for studying on liver cancer from TCM spleen deficiency factors, and spleen-deficiency may be an important factor in the formation of liver cancer and poor prognosis.
