摘 要:目的:观察黛矾散及其各组分对α-奈异硫氰酸脂(ANIT)诱导的大鼠肝内胆汁淤积的影响,以进一步探究黛矾散治疗肝内胆汁淤积的作用机理。方法:将30只雄性Wistar大鼠随机分成正常组、模型组、青黛组、明矾组、黛矾散组、熊去氧胆酸(UDCA)组,用ANIT复制肝内胆汁淤积模型,观察黛矾散及其组分对大鼠肝内胆汁淤积大鼠血清学及肝组织学变化的影响。结果:与正常组大鼠比较,模型组大鼠血清ALT、TBil、DBil、ALP、TBA显著升高(P<0.05);与模型组比较,黛矾散组及UDCA组大鼠ALT、TBil、DBil、ALP、TBA显著降低(P<0.05),青黛组大鼠ALT显著降低(P<0.05),明矾组大鼠ALP显著降低(P<0.05)。肝组织病理结果显示:正常组大鼠肝细胞排列整齐,无炎症细胞浸润,无胆管增生;模型组大鼠肝小叶结构破坏,门管区大量炎症细胞浸润,胆管轻度增生;各治疗组大鼠肝脏炎症细胞浸润及胆管增生有不同程度减轻。结论:黛矾散及其组分能不同程度拮抗ANIT所致胆管损伤所导致的肝内胆汁淤积。Objective: To observe the effects of Daifansan and its components on ANIT induced intrahepatic cholestasis in rats and its mechanism. Methods: 30 male rats were randomly divided into normal group, model group, Qing-dai group, Mingfan group, Daifansan group and UDCA group. Ntrahepatic cholestasis model was reproduced by ANIT so as to observe the effects of Daifansan and its components on in- trahepatic cholestasis in rats serum and liver tissue. Results: compared with normal group, ALT, TBit, DBil, ALP and TBA in model group significantly higher (P〈0.05); Compared with model group, ALT, TBil, DBil, ALP and TBA in Daifansan group and UDCA group decreased signify-cantly (P〈 0.05), ALT in Qingdai group and ALP in Mingfan group also decl-ined significantly (P〈0.05). The pathological results show that liver cells in normal group order neatly, with no inflammatory cellinfiltration, no bile duct hyperplasia; Lobule structure in model group destructed, with a large number of i-nflammatory cells infiltration in portal area and mild bile duct hyperplasia: Inflammatory cells infiltration and bile duct hyperplasia in treatment group ease to different degree. Conclusion: Daifansan and its components can antagonize intrahepatic cholestasis caused by ANIT induced bile duct injury in different deg-ree.
